Optimization of extraction technology of total flavonoids from Pouzolzia zeylanica(L.)Benn and its effect on stress ability of nematode
Objective To optimize the extraction process of total flavonoids from Pouzolzia zeylanica(L.)Benn by orthogonal design and explore its effect on the anti-stress ability of Caenorhabditis elegans.Methods The content of total flavonoids in Pouzolzia zeylanica(L.)Benn was determined by NaNO2-Al(NO3)3-NaOH color developing method.Using the extraction rate of total flavonoids from Pouzolzia zeylanica(L.)Benn as the indicator,The effects of extraction time,temperature,ratio of solid to liquid and concentration of ethanol on total flavonoids extraction were investigated by single factor and orthogonal design,and the optimal extraction process was selected.By setting up a medication group and a blank group,the effects of total flavonoids on the anti-stress ability of C.elegans were studied by observing the head swing times,H2O2 oxidative stress and anti-heat stress experiments.Results The optimum extraction technology of total flavonoids was as follows:the extraction time was 40 min,the extraction temperature was 70℃,the concentration of ethanol was 60%,and the ratio of solid to liquid was 1∶40(g∶mL).The actual extraction rate of total flavonoids was 1.31%.The number of head swing in the treated group was significantly higher than that in the control group(P<0.01).The mass concentration of total flavonoids in Pouzolzia zeylanica(L.)Benn was 0.300 mg/mL,the average lifespan of the experimental group was prolonged by 32.28%in oxidative stress and 42.96%in heat stress.Conclusion The ultrasonic method is simple,convenient and has high extraction rate.The extracted total flavonoids can enhance the stress ability of C.elegans.
