Study on quality evaluation of Polygalae Radix Praeparata Cum Melle standard decoction based on UPLC characteristic chromatograms and content determination
Objective To establish the UPLC method of Characteristic chromatograms and three components content determination of Polygalae Radix Praeparata Cum Melle standard decoction in order to evaluate the quality of decoction made by Polygalae Radix Praeparata Cum Melle docoction pieces from different origins.Methods The UPLC was used to establish the Characteristic chromatograms and tenuifolin,polygalaxanthone Ⅲ and 3,6′-disinapoyl sucrose content determination method of Polygalae Radix Praeparata Cum Melle standard decoction,and 16 batches of standard decoctions were determined.The high resolution mass spectrometer was used to identify the common peaks and the cluster analysis and partial least squares discriminant analysis(PLS-DA)were used to analyze the common peaks.Results Nine common peaks were determined in the Characteristic chromatograms of Polygalae Radix Praeparata Cum Melle standard decoction while the peaks were identified as sibiricose A5,sibiricose A6,polygalaxanthoneⅢ,3,6′-disinapoyl sucrose,arillanin A,tenuifolin A,tenuifoliose H,tenuifoliose A and 1,2,3,7-tetramethoxyxanthone by high resolution mass spectrometer.The similarities of the 16 batches samples were all greater than 0.900.Cluster analysis and PLS-DA showed there might be differences among the samples from different origins.Conclusion The established Characteristic chromatogram and content determination method of Polygalae Radix Praeparata Cum Melle standard decoction is simple,quick and accurate,which can provide a reference for the quality evaluation of Polygalae Radix Praeparata Cum Melle relative preparations.
