降糖消渴颗粒UPLC指纹图谱及多成分含量测定研究

目的 建立降糖消渴颗粒的UPLC指纹图谱,并测定5种指标成分的质量分数.方法 选用Waters UPLC T3(2.1 mm×150 mm,1.6 μm)色谱柱,以0.1%磷酸-甲醇为流动相梯度洗脱,检测波长为254 nm;流速为0.2 mL/min;柱温为30℃;进样量为1 μL.通过UPLC法建立15批样品的指纹图谱,采用SPSS、SIMCA软件进行统计分析,并对其指认的化学成分进行质量分数测定.结果 共确定21个共有峰.化学计量学的结果将样品分为2类,筛选出5种差异性成分分别为盐酸小檗碱、盐酸巴马汀、盐酸药根碱、黄连碱、表小檗碱.盐酸小檗碱在20.550 0~205.500 0 μg范围内线性关系良好(r=0.999 8),加样回收率为100.68%;盐酸巴马汀在7.690 0~57.654 0 μg范围内线性关系良好(r=0.999 4),加样回收率为99.99%;盐酸药根碱在7.920 0~47.520 0 μg范围内线性关系良好(r=0.999 8),加样回收率为98.81%;黄连碱在8.915 0~66.660 0 μg范围内线性关系良好(r=0.999 8),加样回收率为99.01%;表小檗碱在8.108~60.810 0 μg范围内线性关系良好(r=0.999 8),加样回收率为98.87%.结论 建立的降糖消渴颗粒指纹图谱及含量测定方法稳定可靠,为降糖消渴颗粒的质量评价提供了科学参考.
Study on UPLC fingerprints and multi-component content determination of Jiangtang Xiaoke Granules
Objective To establish UPLC fingerprints of Jiangtang Xiaoke Granules and a method for the content determination of 5 index components.Methods The chromatographic column Waters UPLC T3(2.1 mm×150 mm,1.6 μm)was selected with 0.1%phospho-methanol as mobile phase by gradient elution,and the detection wavelength was 254 nm.The flow rate was 0.2 mL/min.The column temperature was 30℃,and the injection volume was 1 μL.The fingerprints of 15 batches of samples were established by UPLC method.SPSS and SIMCA software were used for statistical analysis,and the content of chemical components was determined.Results A total of 21 common peaks were identified.The chemometrics results divided the samples into 2 categories and screened out 5 different components:Berberine hydrochloride,Palmatine hydrochloride,Jatrorrhizine Hydrochloride,Coptisine and Epiberberine.Berberine hydrochloride had a good linear relationship between 20.550 0-205.500 0 μg(r=0.999 8)and the average recovery rate was 100.68%.Palmatine hydrochloride had a good linear relationship between 7.690 0-57.654 0 μg(r=0.999 4)and the average recovery rate was 99.99%.Jatrorrhizine hydrochloride had a good linear relationship between 7.920 0-47.520 0 μg(r=0.999 8)and the average recovery rate was 98.81%.Coptisine had a good linear relationship between 8.915 0-66.660 0 μg(r=0.999 8)and the average recovery rate was 99.01%.Epiberberine had a good linear relationship between 8.108 0-60.810 0 μg(r=0.999 8)and the average recovery rate was 98.87%.Conclusion The established fingerprint of Jiangtang Xiaoke Granules and its content determination method are stable and reliable,which can provide scientific reference material for the quality specification of this preparation.
