Study on the difference between Citrus fruit medicinal materials based on chemometrics
Objective To establish an HPLC analysis method for the simultaneous determination of 5 flavonoids include narirutin,naringin,hesperidin,rhoifolin and neohesperidin,and analyze the differences of 82 batches of the five Citrus fruits medicinal materials.Methods DIKMA Platisil column(4.6 mm×250 mm,5 μm)was used,mobile phase was consisted of acetonitrile-0.1%formic acid(15∶85),the detection wavelength was 238 nm,the flow rate was 1.0 mL/min,and the column temperature was 35℃.The differences between species of Citrus fruits medicinal were studied by analysis of variance and chemometrics.Results The results showed that the determination of 5 flavonoids manifested a good liner relationship(r>0.9990),and the average recovery rate was 98.37%-103.24%.The relative standard deviations(RSDs)of precision,stability,and repeatability were all less than 3.0%.Statistical analysis showed that the content of the five flavonoids was significantly different between the five medicinal materials.Chemometrics showed that neohesperidin,hesperidin and naringin were the differential components of the five medicinal materials.Neohesperidin were the main differential components of Aurantii Fructus and Aurantii Fructus Immaturus of Citrus Aurantium,Aurantii Fructus and Aurantii Fructus Immaturus of different origins.Conclusion This study has established a simple,feasible,precise and stable multi-index content determination method for Citrus medicinal materials,which can provide reference for the identification,quality evaluation and clinical application of citrus fruit medicinal materials.
