Evaluation of anti-inflammatory and antioxidant activities of Scutellaria baicalensis extracts based on zebrafish modelin
Objective To explore the toxicity,anti-inflammatory and antioxidant effects of Scutellaria baicalensis extract by using zebrafish models.Methods An injury inflammation model was constructed by truncating the caudal fin of transgenic zebrafish Tg(colo1a:EGFp).The number of green fluorescent protein(EGFp)-labeled neutrophils and macrophages was observed under fluorescence microscope to evaluate the anti-inflammatory cell aggregation and reverse migration ability of Scutellaria baicalensis extract.Using LPS induction to construct an oxidative stress model,DCFH-DA fluorescent probe was used to stain the levels of reactive oxygen species(ROS)in zebrafish to evaluate the antioxidant effect of Scutellaria baicalensis extract on zebrafish.Results Scutellaria baicalensis extract at concentrations of 0.60 and 1.25 mg/mL both inhibited inflammatory cell aggregation and promoted inflammatory cell clearance in a dose-dependent manner.Scutellaria baicalensis extract at concentrations of 0.020 and 0.040 mg/mL reduced the relative fluorescence intensity of zebrafish ROS in a dose-dependent manner.Conclusion Scutellaria baicalensis extract has a notable anti-inflammatory and antioxidant effect.
