Simultaneous determination of four components in Sujie Oral Liquid by UHPLC-MS/MS
Objective To establish a method for the simultaneous and rapid determination of four components,i.e.liquiritin,rosmarinic acid,platycodon D and glycyrrhizic acid in Sujie Oral Liquid by ultra high performance liquid chromatography-tandem quadrupole mass spectrometry(UHPLC-MS/MS),so as to control the quality of Sujie Oral Liquid.Methods The samples were separated on an Agilent Extend C18(2.1 mm×100 mm,1.7 μm)column with a mobile phase containing 0.1%formic acid(A)and acetonitrile(B)by gradient elution.The column temperature was set at 35℃,and the flow rate was 0.3 mL/min.ESI ion source was used.In multiple reaction monitoring mode,they were analyzed in negative ion mode.The ion pairs were as follows:liquiritin 417.1/255.1(135),rosmarinic acid 359/197.1(160.9),platycodon D 1 223.6/681.0,glycyrrhizic acid 821.4/350.9.Results The four components showed good linear relationships within their mass concentration ranges(r>0.999),whose average recoveries were 98.80%-100.84%with the RSDs of 1.82%-2.53%.In 10 batches of Sujie Oral Liquids,the mass concentrations of liquiritin,rosmarinic acid,platycodon D,and glycyrrhizic acid were 0.140 8-0.159 2,0.082 4-0.142 1,0.035 9-0.069 8,and 0.665 9-0.723 8 mg/mL,respectively.Conclusion The method of simultaneous determination for four components in Sujie Oral Liquid is fast,accurate and highly reproducible,which provides experimental basis for the quality evaluation of Sujie Oral Liquid.
