Analysis of chemical composition and determination of multi-component content of Stemona tuberosa Lour standard decoction based on UPLC-Q-TOF-MS
Objective Based on UPLC-Q-TOF-MS technology,the chemical composition of S.tuberosa Lour.standard decoction was systematically analyzed,and a method was established for the determination of the multi-component content of S.tuberosa Lour.standard decoction.Methods The chemical composition was localized and identified by matching the mass spectral information,fragment ion information and retention time.Determination of three chlorogenic acids in S.tuberosa Lour.standard decoction was performed by UPLC.The column was packed with octadecylsilane-bonded silica gel(100 mm×2.1 mm,1.6 µm),with a mobile phase of acetonitrile-0.1%formic acid for gradient elution,at a column temperature of 25℃,a flow rate of 0.25 mL/min,a sample size of 1 μL and a detection wavelength of 210 nm.Results A total of 27 chemical components were identified,including 11 phenolic acid components and 16 alkaloid components.The average contents of neochlorogenic acid,chlorogenic acid and cryptochlorogenic acid in 15 batches of S.tuberosa Lour.standard decoction were 1.674,1.478 and 1.033 mg/g,respectively,and the average transfer rates were 56.90%,61.62%and 49.63%,and there were some differences in the transfer rates between different batches.Conclusion The chemical composition of the standard decoction of S.tuberosa Lour.was resolved by UPLC-Q-TOF-MS,and a method for the determination of its chlorogenic acid,neochlorogenic acid and cryptochlorogenic acid content was constructed by UPLC,which was stable and reliable,with a high degree of precision of the instrument and good reproducibility,so as to provide theoretical basis for the in-depth understanding and utilization of the medicinal value of S.tuberosa Lour.
