Study on the antipyretic mechanism of different components of compound Xiangyu oral liquid
Objective To investigate the antipyretic effect and mechanism of different components of compound Xiangyu oral liquid on dry yeast-induced fever in rats,and explore the internal relationship between the pharmacodynamic material basis,efficacy and mechanism of the antipyretic effect of components of compound Xiangyu oral liquid.Methods The fever model in SD rats was established by subcutaneous injection of dry yeast on the back.The anal temperature of rats was measured in each group in 6 hours after drug interventions.The maximum temperature rise(ΔTmax)and temperature response index(TRI)were calculated.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of cyclic adenosine monophosphate(cAMP),TNF-α,IL-6 in plasma and cAMP and prostaglandin E2(PGE2)in the hypothalamus.Results Compared with the model group,the whole components group,water extract group and ethanol extract group of compound Xiangyu oral liquid significantly reduced the body temperature of rats induced by dry yeast(P<0.01 or P<0.05).The content of cAMP and IL-6 in plasma of rats in whole components group,water extract component group and alcohol extract component group of compound Xiangyu oral liquid decreased significantly(P<0.01 or P<0.05).The content of TNF-α in plasma of rats in whole components group and alcohol extract component group decreased significantly(P<0.01).The cAMP content in hypothalamus of whole components group,water extract component group and alcohol extract component group was significantly decreased(P<0.01 or P<0.05).The content of PGE2 in hypothalamus of compound Xiangyu oral liquid group was clearly decreased(P<0.05).Conclusion The water extract and alcohol extract of compound Xiangyu oral liquid have good antipyretic effect.The antipyretic mechanism of water extract may be related to the decrease of plasma cAMP,IL-6 and hypothalamic cAMP levels.The antipyretic effect of the alcohol extract components may be related to the reduction of cAMP,TNF-α,IL-6 in plasma and cAMP levels in hypothalamus.
