Study on the effect of Yijing Decoction granules on serum metabolomics in mice with premature ovarian failure based on UPLC-TOF-MS
Objective To investigate the effect of Yijing Decoction granules on serum metabolomics in mice with premature ovarian failure by ultra-performance liquid chromatography tandem time-of-flight mass spectrometry(UPLC-TOF-MS).Methods The experimental mice were randomly divided into the model group,the positive control group,the low and high doses of Yijing Decoction granules groups and blank group,6 mice in each group.The mouse model of premature ovarian failure was induced using cyclophosphamide,and the drug was administered by gavage for intervention once a day for 30 days.The changes of endogenous metabolites in mouse serum samples were analyzed by UPLC-TOF-MS.The potential differences of metabolites were screened by the value of the projected importance of the variables(VIP)and the results of t-test by applying the methods of principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA).The metabolic pathway enrichment analysis was performed using the MetaboAnalyst platform.Results Metabolomics analysis showed that there were 74 potentially different metabolites in the model group compared with the blank group,and there were 130 potentially different metabolites in high-dose Yijing Decoction granules group compared with the model group.After re-screening,25 different metabolites were identified,and 11 KEGG pathways were enriched,including histidine metabolism,citric acid cycle,aminosugar and nucleotide sugar metabolism,fructose and mannose metabolism,and lysine degradation,etc.Conclusion The mechanism of the therapeutic effect exerted by Yijing Decoction granules on cyclophosphamide-induced POF mice may be related to the improvement of the levels of differential metabolites in serum,as well as the modulation of histidine,citric acid cycle,fructose and mannose,and lysine metabolism.
