Expression and significance of miR-206,TGF-β1 and Smad3 in placental tissue of patients with hypertensive disorders of pregnancy
Objective To explore the expression and clinical significance of microRNA-206(miR-206),transforming growth factor-β1(TGF-β1)and Sma-and Mad-related protein 3(Smad3)in placental tissue of patients with hypertensive disorders of pregnancy.Methods A total of 60 patients with gestational hypertension(gestational hypertension group),60 patients with preeclampsia(preeclampsia group),and 60 healthy pregnant women in late pregnancy(control group)who were treated at Hebei Petro China Central Hospital from June 2020 to January 2023 were selected as the study subjects.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the mRNA levels of miR-206,TGF-β1 and Smad3,and immunohistochemistry was used to detect the protein expressions of TGF-β 1 and Smad3 in placental tissues.Pearson correlation was applied to analysis the correlation between the levels of miR-206,TGF-β1 mRNA and Smad3 mRNA levels in placental tissues and clinical data in patients with hypertensive disorders of pregnancy.Receiver operating characteristic(ROC)curves were used to analyze the value of postpartum placental tissue miR-206,TGF-β1 mRNA and Smad3 mRNA levels in assessing the condition of pregnant women with hypertensive disorders of pregnancy.Results The diastolic blood pressure,systolic blood pressure,24-hour urine protein,TGF-β1 mRNA,Smad3 mRNA,positive expression rate of TGF-β1 protein,and positive expression rate of Smad3 protein in the control group,gestational hypertension group,and preeclampsia group increased in turn,while albumin,neonatal weight,placental weight,and miR-206 decreased sequentially,with significant differences(F/x2=39.400-834.610,P<0.05).In patients with gestational hypertension,miR-206 in placental tissue was negatively correlated with systolic blood pressure,diastolic blood pressure,and 24 h urine protein,and positively correlated with albumin,neonatal weight,and placental weight(r=-0.514-0.507,P<0.05).TGF-β1 mRNA and Smad3 mRNA were negatively correlated with albumin,neonatal weight and placental weight,and were positively correlated with systolic blood pressure,diastolic blood pressure,and 24 h urine protein(r=-0.512-0.519,P<0.05).The AUC of miR-206,TGF-β1mRNA and Smad3 mRNA levels in placental tissue for alone and in combination evaluating the condition of pregnant women with hypertensive disorders were 0.781,0.840,0.837 and 0.951,respectively.Conclusion MiR-206 is lowly expressed in placental tissues of patients with hypertensive disorders,while TGF-β1 and Smad3 are highly expressed.The three may be involved in the progression of hypertensive disorders.
