摘要
目的 应用生物信息软件分析细粒棘球蚴14-3-3 (Eg14-3-3)蛋白氨基酸序列、二级结构、三级结构并预测B细胞表位和T细胞表位,为后续确定和筛选优势抗原表位、研制安全高效的多肽疫苗及特异性诊断抗原奠定基础. 方法 采用DNAstar软件、ProPred MHC Class-Ⅱ Binding Peptide Prediction Server和Biosun等软件对Eg14-3-3的序列、结构及B细胞表位和T细胞表位进行预测. 结果 Eg14-3-3为相对分子质量27 940、含有247个氨基酸的酸性蛋白质,其二级结构主要由有6个α-helix结构中间连接短β-折叠及不规则卷曲结构,序列具有高度多变性,预测该蛋白存在14个B细胞表位区域和6个T细胞表位. 结论 分析了蛋白氨基酸序列、二级结构、三级结构,并预测出Eg14-3-3存在14个B细胞表位和6个T细胞表位.
Abstract
Objective To analyze and predict the animo acid sequence,secondary and tertiary structure,and B cell and T cell epitopes of Echinoccocus granulosus protein 14-3-3 with bioinformatics tools for screening epitopes vaccine and diagnostic antigen.Methods The protein secondary structure was analyzed by DNAstar,and B cell and T cell epitopes of Eg14-3-3 were predicted by DNAstar,Biosun software and Propred MHC class-Ⅱ Binding Peptide Prediction Server software,the tertiary structure was predicted with Swiss-model and Rasmol software.Results Eg14-3-3 is a kind of acid protein consisting of 247 amino acids,with Mr 27 940.The secondary structure containing 6 α-helix linked with short β-sheet and coil,the sequence has high variability.14 B cell epitopes and 6 T cell epitopes were predicted.Conclution The study analyzed the sequence,secondary structure and tertiary structure of Eg14-3-3 and 14 B cell epitopes and 6 T cell epitopes predicted.
