Post-translational modification(PTM)are important epigenetic mechanisms in the regulation of various cellular bio-logical processes,playing a significant role in immune regulation and the maintenance of homeostasis.Lactate metabolism and lactate-mediated modification of histone through lactylation are involved in the pathophysiological processes of several lung diseases.Understanding the mechanism of action of PTM in lung diseases is crucial for the discovery of new therapeutic targets and contributes to a better understanding of the development of lung diseases.This review summarizes the mechanisms and pathways of lactate metabo-lism and lactation modification in both oncological and non-oncological respiratory diseases.It also describes potential therapeutic tar-gets and aims to investigate the relevance of lactation modification in lung diseases,to develop therapeutic strategies that could facili-tate the translation of this research from the laboratory to clinical settings.
