生物样品中聚苯乙烯纳米塑料指示物的热裂解-气相色谱质谱检测方法研究

目的 采用热裂解-气相色谱质谱法(Py-GC/MS)建立一种从生物组织样品中定量测定纳米级聚苯乙烯(PS)塑料的方法.方法 通过比较和优化不同消解液、蛋白沉淀剂以及仪器检测参数体系,确定生物样品中 PS 塑料的最优前处理和检测方法.通过开展实验研究确定方法特性指标(包括检出限、定量限、精密度和准确度),并基于方法开展了实际样品测定.结果 本研究建立了大鼠肝和肺组织样本经 10%氢氧化钾溶液消解和无水乙醇蛋白沉淀后上 Py-GC/MS 检测的方法,PS 在0.05~2.00 μg范围内线性关系良好,相关系数为 0.999,检出限和定量限分别为 0.012 和 0.036 μg/g,且当添加水平为低浓度(2 μg/g)、中浓度(10 μg/g)、高浓度(30 μg/g)PS时,其加标回收率分别为 68.0%~85.0%、80.0%~105%和 62.0%~93.1%,精密度(RSD)范围在 7.8%~15.2%.结论 本研究建立的方法满足方法学要求,且为其他生物样本及其他类型微塑料的检测提供了参考,为进一步开展纳米塑料的人群内暴露水平研究和健康风险评估奠定了技术基础.
Determination of polystyrene nanoplastics indicator in biological samples by pyrolysis-gas chromatography/mass spectrometry
Objective To establish a method for the quantitative determination of polystyrene(PS)nanoplastics in biological tis-sue samples by pyrolysis-gas chromatography/mass spectrometry(Py-GC/MS).Methods Different digestion solutions,protein pre-cipitants,and instrument detection parameter systems were compared and optimized to determine pretreatment and determination meth-od for PS plastics in biological samples.We determined the method characteristic indicators(including detection limit,quantification limit,precision and accuracy)by conducting experimental studies,and carried out actual sample measurements based on the method.Results In this study,a Py-GC/MS method was established for the detection of rat liver and lung tissue samples after digestion with 10%potassium hydroxide solution and precipitation of anhydroethanol protein.PS had a good linear relationship within the range of 0.05-2.00 μg,with a correlation coefficient of 0.999,and the limit of detection and the limit of quantification were 0.012 and 0.036 μg/g,respectively.When the spiking level of PS was low(2 μg/g),medium(10 μg/g),or high(30 μg/g),the recovery rates were 68.0%-85.0%、80.0%-105%and 62.0%-93.1%,respectively,and the relative standard deviation ranged from 7.8%to 15.2%.Conclusion The method established in this study meets the methodological requirements and can provide a reference for the detection of other biological samples and other types of microplastics,laying a technical foundation for further population exposure level and health risk assessment of nanoplastics.
