阿维链霉菌高产菌株的复合筛选

目的 筛选阿维链霉菌高产菌株.方法 采用ARTP诱变和aveR基因过表达2种选育方式对阿维链霉菌进行复合筛选.结果 经ARTP诱变筛选出RW-51诱变菌株效价为7 268 U/mL,高于原始菌株8.9%.再结合aveR基因过表达菌株的构建,筛选出高产菌株RW-997初筛效价7 649 U/mL,高于RW-51诱变菌株7.7%,高于原始菌株14.6%.结论 筛选出1株高产菌株RW-997,用于大生产.
Compound Screening of High-yield Strain of Streptomyces avermitilis
Objective The purpose of this study is to screen high-yield strains of Streptomyces avermitilis.Methods In this study,two breeding methods,ARTP mutation and aveR gene overexpression,were used to screen Streptomyces avermitilis.Results The RW-51 mutant strain used in this investigation had a titer of 7 268 U/mL,which was 8.9%greater than the original strain after ARTP mutagenesis screening.When the aveR gene was constructed and overexpressed,a high-yield strain RW-997 was screened with a titer of 7 649 U/mL,which was 14.6%greater than that of the original strain and 7.7%higher than that of the mutant strain RW-5.Conclusion One high-yielding strain RW-997 was selected for large-scale production.
