Optimization of Carbon and Nitrogen Sources and Trace Elements for Streptomyces kanamyceticus Production
The growth and reproduction medium of Streptomyces kanamyceticus was optimized by a series of single-factor and multi-factor experiments. The results showed that the main factors affecting the growth of the solid medium of Streptomyces kanamyceticus were the choice of carbon source and the pH adjustment before the medium sterilization. The experiment found that caramel was the optimum carbon source, and that the pH of the medium was approximately 7.50 prior to medium sterilization. The choice of carbon source and the inclusion of trace elements are the primary factors determining the accumulation of metabolites in the Streptomyces kanamyceticus fermentation medium. The experimental results are as follows:the best carbon source is lactose, followed by the filtration of fermentation broth after adding 0.03%FeSO4 to the medium. The speed was increased by 65.0%compared with the control, and the potency of the shake flask was increased by 21.97%compared with the control.
