Optimization of Fermentation Process of Dodecarbon Acid bacteria strains
Objective To improve the acid production of Candida tropicalis by optimizing the fermentation process.Methods The effects of seed age,fermentation culture temperature,shake flask rotation speed,inoculation volume,filling volume,and sources for the 2 g/L of sucrose,the ideal nitrogen source for 3 g/L extract yeast,bacteria fermentation at 28 ℃,and fermentation pH value on acid production were also studied.The carbon source,nitrogen source,and other components in the fermentation medium were optimized.Results Fermentation to develop the optimal carbon 200 r/min,25 mL/250 mL bottle,with a 15%seed volume,under the condition of fermentation pH value to 7.0~7.5,the average yield of dodecarbonate can reach 130 g/L.Conclusion The carbon and nitrogen supply components of the fermentation medium were improved,along with the fermentation process,resulting in an increase in the fermentation acid production to 130 g/L.At the same time,The addition of sodium chloride and magnesium sulfate in the fermentation medium is not conducive to the production of acid by the strains.
