ATP-柠檬酸裂解酶研究进展

文章从亚细胞定位、结构、酶学活性、作用机制、功能等方面介绍了近年来ATP-柠檬酸裂解酶(ACL)的研究进展.ACL酶的结构复杂,不同的生物体中其结构不同;研究发现,随着进化的发生,该酶的两个异源结构有向同源结构方向发展的趋势.ATP-柠檬酸裂解酶在生物体中发挥重要的作用,如:生成胞间乙酰-CoA、作为脂肪酸合成的调控酶、提高植物抗逆性、参与有性生殖的调节等.近年来,对ACL的研究主要集中在酶学活性上,发现其能提高油菜的产油量和抗旱性.今后可对各种作物的ACL功能及开发利用进行深入研究,尤其在提高作物产油量和抗旱性方面应加大研究力度,提高产油作物的产油量、增强作物对干旱环境的抵抗力;此外,利用不断发展的基因组学、蛋白质组学和宏基因组学等现代分子生物学技术和先进、有效的手段,深入发掘ACL的潜在功能,扩大ACL在各个领域的应用.
Advances in ATP-citrate lyase research
Advances in ATP-citrate lyase (ACL) research in recent years were described from the aspects of subcellular localization,structure,enzyme activities,action mechanism and function.The structure of ACL is especially complex,which varies in different organisms.It has been found that two heterologous structures of the enzyme tend to develop to homologous structure with evolution.ATP-citrate lyase plays a key role in organisms.There are four main functions of ACL including generating intracellular acetyl-CoA,regulating enzymes of fatty acid synthesis,improving plant resistance and regulating sexual reproduction.Relevant ATP-citrate lyase research in recent years focused mostly on enzyme activities ACL could increase the oil content and drought resistance of oilseed rape.The functional studies of ATP-citrate lyase,in particular improving crop oil production and crop drought resistance,will be the future research emphasis.Potential functions of ACL should be developed using genomics,proteomics and metagenomics to expand ACL application in every field.
