Advances in ATP-citrate lyase (ACL) research in recent years were described from the aspects of subcellular localization, structure, enzyme activities, action mechanism and function. The structure of ACL is especially complex, which varies in different organisms. It has been found that two heterologous structures of the enzyme tend to develop to homologous structure with evolution.ATP-citrate lyase plays a key role in organisms. There are four main functions of ACL including generating intracellular acetyl-CoA,regulating enzymes of fatty acid synthesis, improving plant resistance and regulating sexual reproduction.Relevant ATP-citrate lyase research in recent years focused mostly on enzyme activities ACL could increase the oil content and drought resistance of oilseed rape. The functional studies of ATP-citrate lyase,in particular improving crop oil production and crop drought resistance, will be the future research emphasis.Potential functions of ACL should be developed using genomics, proteomics and metagenomics to expand ACL application in every field.
