[Objective]The present experiment was conducted to optimize microwave-enzyme synergistic extraction process of polysaccharide from Sarcodon imbricatus in Ganzi,Sichuan,in order to provide new extraction process for improve extraction rate of polysaccharide from S.imbricatus.[Method]Using S.imbricatus from Ganzi,Sichuan as materials and extraction rate of polysaccharide as indexes,the polysaccharide were extracted from S.imbricatus by microwave-enzyme synergistic method,based on single factor experiment,and the process conditions were optimized using Box-Behnken central complex design and response surface analysis method.[Result]The results showed that,some factors affecting extraction rate of polysaccharide from S.imbricatus were ranked,as follows:complex enzyme dosage>enzymolysis temperature>microwave power>microwave time.Furthermore,the complex enzyme dosage and enzymolysis temperature had extremely significant effects on extraction rate of polysaccharide(P<0.01),and the interaction between enzymolysis temperature and microwave time had significant effects on extraction rate of polysaccharide(P<0.05).In addition,the optimum process conditions were as follows:cellulase and papaya proteinase at mass ratio of 1∶1 in complex enzyme,complex enzyme dosage of 0.70%,pH 5.0,microwave time of 4.3 min,microwave power of 550 W,enzymolysis temperature of 51 ℃.Under the above optimum conditions,the extraction rate of polysaccharide was 16.09%,and the model predicted value was 16.2537%,thus the relative error was 1.02%.[Conclusion]The established model can effectively predict extraction efficiency of polysaccharide from S.imbricatus,and the optimized process parameter can be also applied in actual production.
