摘要

[目的]明确山东省不同地区灰飞虱种群对新型介离子类杀虫剂三氟苯嘧啶的敏感性,探讨亚致死剂量三氟苯嘧啶对灰飞虱保护酶和代谢酶活性的影响,为田间科学、合理使用三氟苯嘧啶防治灰飞虱提供理论依据.[方法]分别采用稻苗浸渍法和点滴法测定三氟苯嘧啶对山东6个灰飞虱种群[鱼台(水稻)种群、鱼台(小麦)种群、济阳种群、岱岳种群、莱芜种群和长清种群]3龄若虫和成虫的室内活性,并比较亚致死剂量(LC10和LC30)三氟苯嘧啶处理对灰飞虱3龄若虫3种保护酶[超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)]和3种解毒酶[谷胱甘肽S-转移酶(GSTs)、羧酸酯酶(CarE)和细胞色素P450酶(P450)]活性的影响.[结果]稻苗浸渍法结果显示,三氟苯嘧啶对灰飞虱3龄若虫的致死中浓度(LC50)介于0.776~1.644 mg/L,对成虫的LC50介于0.752~1.076 mg/L.点滴法测定结果显示,三氟苯嘧啶对灰飞虱3龄若虫的致死中量(LD50)介于3.487~7.441×10-4 µg/头,对成虫的LD50介于1.947~4.196×10-4 µg/头.酶活性测定结果显示,与对照相比,三氟苯嘧啶LC10和LC30 2个亚致死剂量处理72 h后,灰飞虱体内SOD、POD、GSTs和P450活性明显升高,但CarE活性显著降低(P<0.05,下同),且呈明显的剂量效应;三氟苯嘧啶低剂量处理(LC10)能显著诱导CAT活性升高,高浓度处理(LC30)能抑制CAT活性.[结论]三氟苯嘧啶对灰飞虱具有较好的触杀和胃毒活性,山东省不同地区灰飞虱种群对三氟苯嘧啶的敏感性较高.灰飞虱可通过诱导POD、SOD、GSTs和P450活性升高来抵抗或减轻三氟苯嘧啶的毒害.

Abstract

[Objective]In order to provide useful information in the application of trifluorophenyl for the management of L.striatellus in the field.The different sensitivities of six Laodelphax striatellus Fallén populations collected from in Shandong,China to trifluorophenyl(a new mesoionic insecticide)were detected.The effects of the sublethal doses(LC10 and LC30)of trifluorophenyl on the activities of protective and detoxifying enzymes in adults of L.striatellus were also as-sessed.[Method]The bioactivities of trifluorophenyl to the 3rd instar nymphs and adults of six populations collected from'Daiyue,Laiwu,Changqing,Jiyang and Yutai(from wheat and rice,respectively),was evaluated by rice seedlings dipping method and topical application method in the laboratory,respectively.The sublethal effects(LC10 and LC30)of trifluoro-phenyl on the activities of three protective enzymes[superoxide dismutase(SOD)、peroxidase(POD)and catalase(CAT)]and three detoxifying enzymes[glutathione S-transferase(GSTs)、carboxylesterase(CarE)and cytochrome P450(P450)]in 3rd instar nymphs of L.striatellus tby using seedlings dipping method were also measured.[Result]The results showed that the LD50 values of trifluorophenyl to 3rd instar nymphs of six populations of L.striatellus ranged from 3.487×10-4 µg/insect to 7.441×10-4 µg/insect and to adults from 1.947×10-4µg/insect to 4.196×10-4µg/insect by using topi-cal application method.The LC50 values of trifluorophenyl to 3rd instar nymphs of six populations of L.striatellus ranged from 0.756 mg/L to 1.523 mg/L and to adults from 0.752 mg/L t to 1.076 mg/L by using rice seedlings dipping method.Af-ter 72 h of treatment with two sublethal doses of trifluoropyrimidine,compared with the control group,the activities of POD,SOD,GSTs and P450 in L.striatellus adults were significantly increased,while activities of CarE were significantly inhibited,and showed obvious dose effect.The activities of CAT were increased in low concentrations(LC10)treatment and decreased in high concentrations(LC30)treatment,respectively.[Conclusion]The results indicated that trifluorophe-nyl has high insecticidal activities in contact and stomach toxicity against L.striatellus.The sensitivity of six L.striatellus populations to trifluoropyrimidine was higher,and no sensitive differences in different areas of Shandong province.The protective(POD and SOD)and detoxifying(GSTs and P450)enzymes played important roles in the detoxification and resistance of of L.striatellus to trifluoropyrimidine.These results can be used as the relative sensitivity baseline of L.stri-atellus to trifluoropyrimidine in Shandong province,and will help to provide the valuable information for the application of trifluoropyrimidine in the field.
