归芪益母复方制剂对产后奶牛血液氧化脂质的调节作用

摘要
[目的]探究归芪益母复方制剂对产后奶牛血液氧化脂质的调节作用,以期进一步明确归芪益母复方制剂抗炎和抗氧化的作用机制,为归芪益母复方制剂在产后奶牛保健与疾病防治中的应用提供参考依据.[方法]选取体况相近、2~3胎次的健康待产荷斯坦奶牛15头,将其随机分成对照组(C组,n=7)和试验组(G组,n=8),试验组奶牛产后灌服归芪益母复方制剂500 mL/头,连续灌服6 d,对照组奶牛每日灌服等体积饮用水.产后7 d晨饲前于尾静脉采集血液,制备血浆样本,采用超高效液相色谱-质谱(UPLC-MS/MS)联用技术定性定量检测2组奶牛血浆中氧化脂质含量,分析2组奶牛血浆氧化脂质代谢轮廓的变化规律,筛选产后奶牛血液中的差异氧化脂质并进行代谢通路富集分析.[结果]2组奶牛血浆中共鉴定出44种氧化脂质,其中花生四烯酸及其代谢物18个、亚麻酸及其衍生物12个、二十碳五烯酸及其衍生物9个、二十二碳六烯酸及其衍生物4个和二十二碳五烯酸及其衍生物1个.产后7 d,试验组奶牛血浆氧化脂质代谢轮廓较对照组发生显著变化,8种氧化脂质发生显著变化(P<0.05,VIP≥1,FC≥2或FC≤0.5),其中5种脂质分子显著上调,包括12-HETE、11-HETE、15-HETE、5-HETE和15-oxoETE;3种脂质分子显著下调,包括20-HETE、8,9-EET和TXB2.通路富集分析结果表明,差异脂质分子主要富集在花生四烯酸代谢通路、TRP通道的炎症介质调节通路和PPAR信号通路.[结论]归芪益母复方制剂主要通过花生四烯酸代谢通路、TRP通道的炎症介质调节和PPAR信号通路来调节产后奶牛脂质代谢,从而发挥抗炎和抗氧化作用.
Abstract
[Objective]To investigate the regulation of oxidized lipids in the blood of postpartum dairy cows by Quiqi yimu compound preparation,with a view to further clarifying the anti-inflammatory and antioxidant mechanisms of Quiqi Yimu compound preparation,and to provide a reference basis for the application of Quiqi yimu compound preparation in the health care and disease control of postpartum dairy cows.[Method]Fifteen healthy Holstein cows with similar body condition and 2-3 parities were selected and randomly divided into the control group(group C,n=7)and the experimen-tal group(group G,n=8).The cows in the experimental group were dosed with Guiqi Yimu compound formulation 500 mL/cow after parturition for 6 consecutive d,while the cows in the control group were dosed with an equal volume of drinking water every day.Blood was collected from the tail vein before morning feeding 7 d postpartum and plasma samples were prepared.Ultra performance liquid chromatography-mass spectrometry(UPLC-MS/MS)was used to quali-tatively and quantitatively detect the content of oxidized lipids in the plasma of two groups of dairy cows,analyze the changing law of the metabolic profile of oxidized lipids in the plasma of two groups of dairy cows,and screen for the dif-ferences of oxidized lipids in the blood of postpartum dairy cows and carry out the analysis of metabolic pathway enrich-ment.[Result]A total of 44 oxidized lipids were identified in the plasma of 2 groups of cows,including 18 arachidonic acids and its metabolites,12 linolenic acids and its derivatives,9 eicosapentaenoic acids and its derivatives,4 docosa-hexaenoic acids and its derivatives,and 1 docosapentaenoic acid and its derivatives.At 7 d postpartum,plasma oxidized lipid metabolic profiles of cows in the experimental group changed significantly compared with the control group,with eight oxidized lipids significantly changed(P<0.05,VIP≥1,FC≥2,or FC≤0.5),of which five oxidized lipids molecules were significantly up-regulated,including 12-HETE,11-HETE,15-HETE,5-HETE,and 15-oxoETE,and three were significantly down-regulated,including 20-HETE,8,9-EET,and TXB2.Pathway enrichment analysis showed that the differential lipid molecules were mainly enriched in the arachidonic acid metabolic pathway,the inflammatory mediator-regulated pathway of TRP channels,and the PPAR signaling pathway.[Conclusion]Guiqi Yimu compound preparation mainly regulates lipid metabolism in postpartum cows through arachidonic acid metabolic pathway,inflammatory media-tor regulation of TRP channels,and PPAR signaling pathway,thus exerting anti-inflammatory and antioxidant effects.
