[目的]对文冠果(Xanthoceras sorbifolium)侧生器官边界域(LBD)基因家族成员进行鉴定,并分析其在盐胁迫条件下的表达情况,为文冠果抗逆品种选育及其在盐碱地的推广利用提供理论依据。[方法]以文冠果全基因组数据为基础,从文冠果基因组数据库中鉴定获得LBD基因家族成员,通过ExPASy、MEME等生物信息学工具对XsLBD家族蛋白的理化性质、保守基序、顺式作用元件、基因结构和系统发育关系进行预测分析。对2个月苗龄的文冠果苗进行NaCl浓度为50、100、150 mmol/L盐胁迫处理,以NaCl浓度为0 mmol/L为对照组(CK),对盐胁迫下文冠果根和叶组织中XsLBD家族基因的表达模式进行实时荧光定量PCR检测。[结果]从文冠果基因组中共鉴定到43个基因家族成员,命名为XsLBD1~XsLBD43。XsLBD家族蛋白编码138~404个氨基酸残基,理论等电点(pI)为4。60~9。75,分子量为15696。60~44476。88 kD,亚细胞定位结果显示,除XsLBD4、XsLBD8、XsLBD9、XsLBD20蛋白外,其余所有蛋白均定位于细胞核。有42个XsLBD家族基因分布在13条染色体上,XsLBD43基因则游离在染色体外。基于文冠果、拟南芥、水稻和毛果杨LBD氨基酸序列相似性构建系统发育树,结果显示,178个LBD家族蛋白可分为两大组(Class I和Class II),其中Class I中包含38个XsLBD家族蛋白。XsLBD基因家族成员的外显子数目为1~5个,内含子数目为0~4个;所有XsLBD家族蛋白均含有Motif 2和Motif 3。XsLBD家族基因启动子中包含激素(水杨酸、脱落酸、生长素等)、应激(低温、干旱诱导等)及植物生长发育(种子特异性调控和昼夜节律控制等)响应元件。对文冠果苗进行不同浓度NaCl盐胁迫处理,结果显示,与CK相比,文冠果根、叶13个XsLBD家族基因中相对表达量存在显著差异(P<0。05,下同)的基因分别有12和11个,其中,XsLBD42基因在根和叶中相对表达量较CK高出约40倍。[结论]筛选获得43个XsLBD基因家族成员,其结构相对简单,具有一定保守性;XsLBD家族基因参与调控文冠果生长发育、新陈代谢和应激反应等过程;XsLBD9、XsLBD11、XsLBD23和XsLBD42等基因在文冠果盐胁迫响应中可能发挥重要作用。
Identification of LBD gene family in Xanthoceras sorbifolium and its response to salt stress
[Objective]The members of the lateral organ boundary domain(LBD)gene family of Xanthoceras sorbi-folium were identified and their expression analysis under salt stress conditions were analyzed,which provided theoretical basis for the breeding of stress-resistant varieties of X.sorbifolium and its promotion and utilization in saline-alkali land.[Method]Based on the complete genome data of X.sorbifolium,the members of LBD gene family were identified from the X.sorbifolium genome database.The physicochemical properties,conserved motifs,cis-acting elements,gene struc-ture and phylogenetic relationships of the XsLBD family proteins were predicted and analyzed using bioinformatics tools such as ExPASy and MEME.The 2-month-old seedlings of X.sorbifolium were treated with NaCl concentrations of 50,100 and 150 mmol/L,with 0 mmol/L NaCl as the control group(CK).The expression patterns of XsLBD family genes in the roots and leaves of X.sorbifolium under salt stress were detected by real-time fluorescence quantitative PCR.[Result]A total of 43 gene family members were identified from X.sorbifolium genome,and were named XsLBD1-XsLBD43.XsLBD family proteins encoded 138-404 amino acid residues,with a theoretical isoelectric point(pI)of 4.60-9.75,mo-lecular weight of 15696.60-44476.88 kD,and subcellular localization results showed that all but XsLBD4,XsLBD8,XsLBD9 and XsLBD20 proteins were localized in the cell nucleus.There were 42 XsLBD family genes distributed on 13 chromosomes,while XsLBD43 gene was located outside of the chromosomes.Based on the amino acid sequence similari-ties of LBD from X.sorbifolium,Arabidopsis thaliana,Oryza sativa and Populus tomentosa,the 178 LBD family pro-teins were divided into 2 major groups(Class I and Class II),with 38 XsLBD family proteins in Class I.The exon num-ber of XsLBD gene family members ranged from 1 to 5,while the intron number ranged from 0 to 4.All XsLBD family proteins contained Motif 2 and Motif 3.The promoters of XsLBD family genes contained response elements to hormones(salicylic acid,abscisic acid,auxin),stresses(cold,drought induction),and plant growth and development(seed-specific regulation and circadian rhythm control).Among the 13 XsLBD family genes screened out,12 and 11 genes showed significant differences(P<0.05,the same below)in relative expression levels in roots and leaves compared with CK after different concentrations of NaCl salt stress treatment respectively.Among them,the relative expression level of XsLBD42 gene in roots and leaves was about 40 times higher than that of CK.[Conclusion]A total of 43 XsLBD gene family members were screened,which have relatively simple structures and certain conservativeness;XsLBD family genes are involved in regulating the growth and development,metabolism and stress response of X.sorbifolium;XsLBD9,XsLBD11,XsLBD23 and XsLBD42 may play an important role in the salt stress response of X.sorbifolium.
