Correlation study of SNHG10 low expression with prognosis and drug resistance in ovarian cancer
Objective:To investigate the relationship between long non-coding small nucleolar RNA host gene 10(SNHG10)with cell proliferation,drug resistance,and the prognosis of ovarian cancer(OC)cells.Methods:A total of 88 normal ovarian tissues and 426 OC tissues for differential expression of SNHGs by open data were screened to analyze their correlation with the prognosis of OC patients and evaluate the value of SNHGs in pre-dicting paclitaxel and platinum drug resistance in OC by receiver operating characteristic(ROC)curves.Reverse transcription-quantitative PCR(RT-qPCR)was used to measure the SNHG10 expression level in paclitaxel/carbo-platin-resistant cells(SKOV3-R/SKOV3-CBP)and their parental cells(SKOV3)of OC.Cell lines overexpress-ing SNHG10 were constructed via lentiviral infection in SKOV3 cells,and were divided into a control group(S-eGFP group)and an overexpression group(S-SNHG10 group).Cell proliferation was detected by cell counting kit-8(CCK-8)and colony formation assay.The cell sensitivity to paclitaxel was assessed by CellTiter-Glo lumi-nescent cell viability assay.Results:SNHG10 was significantly underexpressed in OC tissues(P<0.01).Its low expression was significantly associated with poor prognosis of OC patients(P<0.05),and could potentially pre-dict resistance to paclitaxel and platinum chemotherapy(AUC>0.6,P<0.05).Compared with the S-eGFP group,cells in the S-SNHG10 group showed decreased proliferation and enhanced sensitivity to paclitaxel(P<0.001).Conclusion:Overexpression of SNHG10 significantly inhibits the proliferation of OC cells and increases the sensitivity of OC cells to paclitaxel.
