Effect of inner-heat acupuncture for the intervention of rats model with lumbar disc herniation and its potential mechanism
Objective To investigate the effect of inner-heat acupuncture for the intervention of rats model with lumbar disc herniation(LDH)and its potential mechanism.Methods A total of 40 rats were randomly assigned to normal group,model group,inner-heat acupuncture group,or electric acupuncture group,with 10 rats in each group.Except for the normal group,LDH model was established in the remaining 3 groups.After modeling,rats in the inner-heat acupuncture and electric acupuncture groups received inner-heat acupuncture and electric acupuncture for treatment,respectively,whereas neither the normal group or the model group received any intervention.After 2 treatment courses of intervention,pathological changes of dorsal root ganglion tissues were observed in rats of various groups.Thermal paw-withdrawal latency(TPWL),nerve conduction velocity,serum interleukin(IL)-6 and IL-1β levels,tachykinin receptor(NK)1 and NK2 protein expressions in dorsal root ganglion tissues were detected in rats of various groups.Results Rats in the model group presented as swelling neurons and significant cell-apoptosis neurons,irregular-change and nucleolus-ambiguity in cell nucleus,disordered and cavity-form Nissl body in cytoplasm,whereas rats in the inner-heat acupuncture and electric acupuncture groups obtained ameliorated changes as above.Compared with the normal group,rats in the remaining groups obtained decreased TPWL,decreased sensory and motor nerve conduction velocity,elevated serum IL-6 and IL-1β levels,and elevated protein expressions of NK1 and NK2 in dorsal root ganglion tissues.The model group,the electric acupuncture group,and the inner-heat acupuncture group exhibited elevated TPWL successively,increased sensory and motor nerve conduction velocity,decreased serum IL-6 and IL-1β levels,and decreased protein expressions of NK1 and NK2 in dorsal root ganglion tissues(P<0.05).Conclusion Inner-heat acupuncture can relive LDH rats model's pain,improve their sensory and motor nerve conduction velocity.Its mechanism may be related to the inhibition of NK1/NK2 expression and thereby relieving inflammatory responses.
