DNM3OS expression in glioblastoma and its clinical significance based on bioinformatics:an anlytic study
Objective To analyze the expression of dynamin 3 opposite strand/antisense RNA(DNM3OS)in glioblastoma(GBM)and its clinical significance.Methods(1)The Cancer Genome Atlas(TCGA)database and Chinese Glioma Genome Atlas database were employed to analyze DNM3OS expression in GBM.The relation of DNM3OS with GBM patients'clinical characteristics was analyzed according to data obtained from TCGA database.The relation of DNM3OS expression with GBM patients'total survival was analyzed by using the COX regression model.Co-expressed genes of DNM3OS with GBM were obtained from the LinkedOmics database.The Pearson correlation analysis was used to analyze the correlation of DNM3OS expression with co-expressed genes expressions.The correlation of DNM3OS with immune cells was analyzed by using markers of 24 immune cells provided by Immunity articles.(2)Clinical samples of GBM patients and patients with cerebral hemorrhage were obtained,and their DNM3OS expressions were analyzed by using the real-time quantitative fluorescent PCR.Results(1)Information of TCGA database revealed that there was differential expression of DNM3OS in multiple cancers,and it exhibited high expression in GBM tissues,as well as DNM3OS expression was related to isocitrate dehydrogenase(IDH)mutation status and gender in GBM patients(P<0.05).The results of survival analysis indicated that compared with the low expression group,the total survival of GBM patients in the high expression group was shorter(P<0.05).The results of Pearson correlation analysis interpreted that the top 20 genes with the strongest correlation with DNM3OS positively correlated with DNM3OS expression(P<0.05).The results of correlation analysis of immune cell infiltration with DNM3OS expression depicted that DNM3OS expression was related to multiple abundance of immune cell infiltration(P<0.05).(2)The results of clinical data interpreted that DNM3OS expression was lower in GBM cerebral tissues than in brain tissues of patients with cerebral hemorrhage(P<0.05).Conclusion DNM3OS exhibits high expressed in GBM relevant data obtained from TCGA database,and its expression is related to IDH mutation status and gender in GBM patients.Patients with high-expressed DNM3OS obtain adverse prognosis,and DNM3OS is related to multiple abundance of immune cell infiltration.However,DNM3OS expression still needs to be further studied and explored in GBM.
