蓝莓是一种新型的保健果品,已被越来越多的国家所重视.以蓝莓当年生带腋芽茎段为组培外植体材料,搭配IAA、CPPU、ZT等激素,探讨其初代培养过程中消毒时间和不同激素浓度对继代培养适宜的培养基组合.试验结果表明:最佳消毒时间为 75%酒精处理 5 min与 0.1%升汞处理 6 min,成活率达 90%;最佳愈伤增殖培养基为WPM+TDZ 0.05 mg/L+IAA 0.1 mg/L,不定芽分化率达 100%;最佳继代培养基为WPM+IAA 0.1 mg/L+ZT 1 mg/L,株高为 2.73 cm,植株生长健壮,增殖系数为 5.6.
Study on Subculture Technology of Blueberry Tissue Culture
Blueberry is a new kind of health fruit,which has been paid more and more attention in many countries.The suitable medium combination of disinfection time and different hormone concentrations for subculture of blueberry in the course of primary culture was studied by using the stem segment with axillary buds as explants with IAA,CPPU,ZT and other hormones.The results showed that the best disinfection time was 5 min with 75% alcohol and 6 min with 0.1% Hg and the survival rate reached 90% .The optimal medium for callus proliferation was WPM+TDZ 0.05 mg/L+IAA 0.1 mg/L,and the adventitious bud differentiation rate reached 100% .The optimal subculture medium was WPM+IAA 0.1 mg/L+ZT 1 mg/L,the average plant height was 2.73 cm,the growth was strong and the proliferation coefficient was 5.6.This study provides experimental basis for the rapid propagation and promotion of blueberry production,and has certain research significance.
