益肾痛泄方对腹泻型肠易激综合征大鼠IL-2、IL-10、TGF-β、IFN-γ表达的影响

[目的]研究益肾痛泄方对腹泻型肠易激综合征模型大鼠小肠组织白细胞介素2(IL-2)、白细胞介素10(IL-10)、转化生长因子β(TGF-β)、γ-干扰素(IFN-γ)表达水平的影响.[方法]将24只Wistar大鼠按随机数字表法分为正常组、模型组、奥替溴铵组、益肾痛泄方组,每组6只,除正常组大鼠外,其余各组均采用4%醋酸灌肠叠加慢性心理应激法复制腹泻型肠易激综合征模型,奥替溴铵组、益肾痛泄方组大鼠连续给药14 d.观察大鼠的一般情况,大鼠处死后,采用苏木素-伊红(HE)染色法观察各组大鼠小肠组织病理变化,采用实时荧光定量聚合酶链式反应(RT-PCR)及蛋白免疫印迹法(Western Blot)分别检测各组大鼠小肠组织IL-2、IL-10、TGF-β、IFN-γ mRNA及蛋白表达水平.[结果]与正常组比较,模型组大鼠小肠出现明显的淋巴细胞灶性浸润,且模型组大鼠小肠组织中IL-2、IL-10、TGF-β、IFN-γ mRNA及蛋白表达水平均显著升高,差异有统计学意义(P<0.01);与模型组比较,奥替溴铵组、益肾痛泄方组大鼠小肠组织未见明显炎症浸润,且小肠IL-2、IL-10、TGF-β、IFN-γ mRNA及蛋白表达水平均显著降低(P<0.05或P<0.01);与奥替溴铵组比较,益肾痛泄方组大鼠小肠组织固有层肠腺丰富,未见明显炎症细胞浸润,IL-2、IL-10、TGF-β、IFN-γ mRNA及蛋白表达水平均低于奥替溴铵组,但差异无统计学意义(P>0.05).[结论]益肾痛泄方可能通过降低IL-2、IL-10、TGF-β、IFN-γ的表达,进而缓解小肠炎症损伤,发挥治疗腹泻型肠易激综合征的作用.
Effects of Yishen Tongxie Formula on the Expression of IL-2,IL-10,TGF-β and IFN-γ in Rats with Diarrhea-type Irritable Bowel Syndrome
[Objective]To study the effects of Yishen Tongxie formula on the expression levels of IL-2,IL-10,TGF-β and IFN-γ in the small intestine of rats with diarrheal irritable bowel syndrome.[Methods]Twenty-four wistar rats were divided into normal group,model group,Otilonium bromide group and Yishen Tongxie formula group by ran-dom number table method,with 6 rats in each group.By using 4%acetic acid enemas combined with chronic psycholog-ical stress,the diarrhea-type irritable bowel syndrome model was built in all groups except the normal group.In the Oti-lonium bromide group and Yishen Tongxie formula group,the related drugs were administered for 14 days after the mod-el was built.The general condition of the rats was observed before execution.HE staining was used to observe the patho-logical changes of small intestine,and RT-PCR and Western blot were used to measure the mRNA and protein expres-sion levels of IL-2,IL-10,TGF-β and IFN-γ in small intestine.[Results]Compared with the normal group,obvious focal lymphocyte infiltration in the small intestine of the rats in the model group was observed,and the mRNA and pro-tein expression levels of IL-2,IL-10,TGF-β and IFN-γ in the small intestine of the rats in the model group were sig-nificantly increased(P<0.01).Compared with the model group,no obvious inflammatory infiltration in small intestinal tissues was observed in Otilonium bromide group and Yishen Tongxie formula group,and the mRNA and protein expres-sion levels of IL-2,IL-10,TGF-β and IFN-γ were significantly decreased in Otilonium bromide group and Yishen Tongxie formula group(P<0.05 or P<0.01).Compared with Otilonium bromide group,abundant intestinal glands in the lamina propria of small intestine and no obvious inflammatory cell infiltration were observed in Yishen Tongxie formula group,and the mRNA and protein expressions of IL-2,IL-10,TGF-β and IFN-γ were lower than those of Otilonium bromide group,with no statistical significance(P>0.05).[Conclusion]Yishen Tongxie formula may alleviate the dam-age of intestinal inflammation and play a role in the treatment of diarrhea-type irritable bowel by reducing the expres-sion of IL-2,IL-10,TGF-β,and IFN-γ.
