清燥养肺方对干燥综合征NOD小鼠Th1/Th2免疫调节的影响

目的:研究清燥养肺方对干燥综合征非肥胖糖尿糖(NOD)小鼠Th1/Th2细胞免疫失衡的干预作用及其相关细胞因子表达的影响,探讨该方治疗干燥综合征的作用机制。方法:将100只雌性干燥综合征NOD小鼠随机分为模型组、硫酸羟氯喹组及清燥养肺方低、中、高剂量组,每组20只。模型组小鼠灌胃给予0。9%氯化钠注射液,硫酸羟氯喹组灌胃给予硫酸羟氯喹溶液(剂量为60 mg/kg),清燥养肺方低、中、高剂量组分别按30 g/kg、60 g/kg、120 g/kg的剂量灌胃给予清燥养肺方药液,灌胃容量均为10 ml/kg。分别于实验第2周、第4周、第6周、第8周,采用湿重法测定小鼠唾液流率;实验8周后股动脉取血,测定各组小鼠血清Th1相关细胞因子[白介素(IL)-2、干扰素(IFN)、肿瘤坏死因子-α(TNF-α)]及Th2相关细胞因子(IL-4、IL-6、IL-10、IL-13)水平。结果:实验第2~6周,各给药组小鼠唾液流率均高于模型组(P<0。05);实验第6周,清燥养肺方高剂量组小鼠唾液流率高于硫酸羟氯喹组(P<0。05);实验第8周,清燥养肺方中、高剂量组小鼠唾液流率高于硫酸羟氯喹组(P<0。05)。与模型组比较,各用药组小鼠血清Th1细胞因子(IFN、TNF-α、IL-2)水平和Th2相关细胞因子(IL-4、IL-6、IL-10、IL-13)水平均明显下降(P<0。05),且清燥养肺方高剂量组IFN、TNF-α、IL-2及IL-4、IL-10、IL-13血清水平均低于硫酸羟氯喹组(P<0。05)。结论:清燥养肺方能够增加干燥综合征NOD小鼠唾液流率,降低其血清Th1/Th2相关细胞因子的水平,可能是通过调节Th1/Th2免疫平衡,抑制细胞因子的促炎效应,从而发挥治疗作用。
Effect of Qingzao Yangfei Decoction on Th1/Th2 immune regulation in NOD mice with Sjogren's syndrome
Objective:To study the intervention effect of Qingzao Yangfei Decoction on Th1/Th2 cell immune im-balance and the expression of related cytokines in Non-obese diabetic(NOD)mice with Sjogren's syndrome,and to ex-plore the mechanism of Qingzao Yangfei Decoction in the treatment of Sjogren's syndrome.Methods:A total of 100 fe-male NOD mice with Sjogren's syndrome were randomly divided into model group,hydroxychloroquine sulfate group and Qingzao Yangfei Decoction low,medium and high dose groups,with 20 mice in each group.The mice in the model group were given 0.9%sodium chloride injection by intragastric administration.The hydroxychloroquine sulfate group was given hydroxychloroquine sulfate solution(60 mg/kg)by intragastric administration.The low,medium and high dose groups of Qingzao Yangfei Decoction were given Qingzao Yangfei Decoction by intragastric administration at 30 g/kg,60 g/kg and 120 g/kg,respectively.The intragastric volume was 10 ml/kg.The salivary flow rate of mice was mea-sured by wet weight method at the 2nd,4th,6th and 8th week of the experiment.After 8 weeks of the experiment,blood was taken from the femoral artery to determine the levels of serum Th1-related cytokines as interleukin(IL)-2,interfer-on(IFN),tumor necrosis factor-α(TNF-α)and Th2-related cytokines(IL-4,IL-6,IL-10,IL-13)in each group.Results:From the 2nd to 6th week of the experiment,the salivary flow rate of mice in each administration group was higher than that in the model group(P<0.05).At the 6th week of the experiment,the salivary flow rate of mice in the high-dose Qingzao Yangfei Decoction group was higher than that in the hydroxychloroquine sulfate group(P<0.05).At the 8th week of the experiment,the salivary flow rate of mice in the medium and high dose groups of Qingzao Yangfei Decoction was higher than that in the hydroxychloroquine sulfate group(P<0.05).Compared with the model group,the levels of serum Th1 cytokines(IFN,TNF-α,IL-2)and Th2-related cytokines(IL-4,IL-6,IL-10,IL-13)in each treat-ment group were significantly decreased(P<0.05),and the levels of IFN,TNF-α,IL-2 and IL-4,IL-10,IL-13 in the high-dose group of Qingzao Yangfei Decoction were lower than those in the hydroxychloroquine sulfate group(P<0.05).Conclusion:Qingzao Yangfei Decoction can increase the salivary flow rate of NOD mice with Sjogren's syndrome and reduce the level of Th1/Th2 related cytokines in serum.It may play a therapeutic role by regulating Th1/Th2 immune balance and inhibiting the proinflammatory effect of cytokines.
