蓝布正提取物对老年痴呆小鼠学习记忆能力的影响及作用机制研究

目的:观察蓝布正提取物(GHE)对老年痴呆(AD)小鼠学习记忆能力的影响并探讨其作用机制。方法:将48只小鼠随机分为正常对照组、模型组、阳性对照组及蓝布正低、中、高剂量组,每组8只。除正常对照组外,余组均采用腹腔注射亚硝酸钠联合皮下注射D-半乳糖制备AD模型小鼠。造模的同时给予相应治疗药物,阳性对照组灌胃给予吡拉西坦溶液(剂量为0。8 g/kg),蓝布正低、中、高剂量组灌胃给予蓝布正提取物溶液(剂量分别为2。5 g/kg、5 g/kg、10 g/kg),灌胃容量均为20 ml/kg。正常对照组和模型组灌胃给予等容量生理盐水。给药2个月后进行Morris水迷宫实验以检测小鼠学习记忆能力,采用免疫组织化学法检测小鼠大脑皮质核因子κB(NF-κB)p65、肿瘤坏死因子α(TNF-α)、磷脂酰肌醇 3-激酶(PI3K)、蛋白激酶B(Akt)的表达水平。结果:与正常对照组比较,模型组逃避潜伏期明显延长(P<0。05),穿越平台次数显著减少(P<0。05),小鼠大脑皮层NF-κB p65、TNF-α、Akt、PI3K表达水平均显著升高(P<0。05);与模型组比较,阳性对照组及蓝布正高、中剂量组小鼠的逃避潜伏期显著缩短(P<0。05),穿越平台次数显著增加(P<0。05);与模型组比较,阳性对照组及蓝布正高、中、低剂量组均能显著降低AD小鼠大脑皮层NF-κB p65、TNF-α、PI3K、Akt的表达水平(P<0。05)。结论:蓝布正提取物能改善AD小鼠的学习记忆能力,其作用机制可能与抑制大脑皮层NF-κB p65、TNF-α、PI3K、Akt的表达水平有关。
Effects of GEI HERBA extract on learning and memory ability of mice with Alzheimer's disease and its mechanism of action
Objective:To observe the effects of GEI HERBA extract on the learning and memory ability of Al-zheimer's disease(AD)mice and study its mechanism of action.Methods:48 mice were randomly divided into 6 groups,which are normal control group,model group,positive control group and GEI HERBA low,medium and high dose groups,with 8 mice in each.Except for the normal control group,the other groups were injected with sodium ni-trite intraperitoneally and D-galactose subcutaneously to replicate AD model mice.The treatment drugs were given orally during the process of modeling.The animals in positive control group were given piracetam solution(0.8 g/kg),and those in the low-,medium-,and high-dose groups were given GEI HERBA extract solution(2.5 g/kg,5 g/kg,and 10 g/kg,respectively).The administrative volume was 20 ml/kg.The mice in the normal control group and the model group were given the same volume of normal saline.After 2 months of treatment,the learning and memory ability was de-termined by Morris water maze,the expression of nuclear factor κB(NF-κB)p65,tumor necrosis factor α(TNF-α),phosphatidylinositol 3-kinase(PI3K),and protein kinase B(Akt)in the cerebral cortex were determined by immunohis-tochemical analysis.Results:Compared with the normal control group,the escape latency of the mice in the model group was significantly prolonged(P<0.05),the number of crossing the platform was significantly reduced(P<0.05),and the expression of NF-κB p65,TNF-α,Akt,and PI3K in the cerebral cortex of mice was significantly increased(P<0.05);compared with the model group,the escape latency of the mice in the positive control group and the high and me-dium dose groups was significantly shortened(P<0.05),and the number of crossing the platform was significantly in-creased(P<0.05);compared with the model group,the expression of NF-κB p65,TNF-α,PI3K,and Akt in the cere-bral cortex of AD mice was significantly reduced in the positive control group and the high,medium,and low dose groups(P<0.05).Conclusion:GEI HERBA extract can improve the learning and memory ability of AD mice,and its mechanism of action may be related to the inhibition of the expression of NF-κB p65,TNF-α,PI3K,and Akt in the ce-rebral cortex.
