Isolation,Identification and Analysis of Biological Characteristics of a Wild Pholiota aurivella
Pholiota aurivella is one of the wild edible and medicinal fungi,which is widely distributed in China,but its exploitation and utilization is limited at present.In order to further develop and utilize the strain resources,a wild edible fungus,No.HC-1,was taken from the willow tree on the campus of Xizang Agricultural and Animal Husbandry College in Linzhi,City,Xizang.The strain was identified as Pholiota aurivella by morphological observation,ITS sequence determination and phylogenetic analysis.The biological characteristics of the optimum growth conditions of the mycelia were studied by single factor test and orthogonal test.The results revealed that for the HC-1 strain of Pholiota aurivella,the optimal carbon source for mycelial growth was sucrose,the best nitrogen source was yeast extract,the optimal pH was 7.0,the preferred temperature was 25°C,the optimal inorganic salt was potassium dihydrogen phosphate,and the most suitable growth factor was vitamin C.Orthogonal experiments indicated that the optimal formula for mycelial growth of strain HC-1 was 25.0 g·L-1 sucrose,3.0 g·L-1 yeast extract,pH 7.0,and 2.0 g·L-1 magnesium sulfate.
