插田泡花色苷的大孔树脂纯化及体外抗氧化活性

采用AB-8 型大孔树脂分离纯化插田泡果实花色苷,并对其抗氧化活性进行了研究.结果显示,采用体积分数70%乙醇为洗脱液,花色苷上样液pH值为 1 时,插田泡花色苷的最佳纯化条件为:动态吸附解吸流速均为1 mL/min,上样液浓度为0.75 mg/mL,用6 倍柱体积的70%乙醇洗脱.纯化前后插田泡花色苷的色价比为1∶15.7,达到了很好的纯化效果.在实验范围内,纯化后的花色苷对DPPH和ABTS+·自由基均有非常强的清除作用,清除率随质量浓度的升高而快速增大,呈正效线性相关;纯化花色苷对 2 种自由基的清除能力大小为ABTS(IC50=0.6349 μg/mL)>DPPH(IC50=0.9044 μg/mL),均远远高于阳性对照VC.结果表明,插田泡纯化花色苷具有十分显著的抗氧化活性.
Purification by Microporous Resin and Antioxidant Activity in vitro of Anthocyanins from Rubus coreanus Maq
The anthocyanins from fresh fruit of Rubus coreanus Maq.was isolated and purified by AB-8 microporous res-in,and its antioxidant activity was studied.Treatment at pH 1 of the sample solution,anthocyanins concentration of the sample solution was 0.75 mg/mL,adsorption and desorption flow rates were 1mL/min and the elution solvent was 6BV of 70%ethanol,a good purification effect was achieved,and the ratio of color value of anthocyanins before and after purification is 1∶15.7.In addition,anthocyanins after purification from fresh fruit of Rubus coreanus Maq.was evaluated for its antioxidant activity.It was found that anthocyanins displayed dose-dependent reducing power,positive linear cor-relation and strong abilities to scavenge DPPH and ABTS free radicals in the concentration range investigated here.And anthocyanins possessed free radical scavenging activity against ABTS(IC50=0.6349 μg/mL)and DPPH(IC50=0.9044 μg/mL)radicals in decreasing order as well as reducing power,the inhibiting abilities were far higher than VC as posi-tive controls.These results show that anthocyanins from Rubus coreanus Maq.have potent antioxidant activity.
