Study on triterpene extraction of Prunella vulgaris L.and hypoglycemic activity in vitro
In this study,Prunella vulgaris L.(P.vulgaris)was used as raw material and three factors(ethanol concen-tration,ultrasonic temperature and ultrasonic power)that had significant effects on the yield of triterpenoids from P.vul-garis were determined by single factor experiment combined with Plackett-Burman(PB)experiment.The response sur-face optimization experiment of the extraction process of triterpenoids from P.vulgaris was carried out by Box-Behnken experimental design method,and the optimal extraction process of triterpenoids from P.vulgaris was obtained.And then,the hypoglycemic activity and anti-inflammatory activity of triterpenoids from P.vulgaris were evaluated.The ex-perimentally proven optimal extraction conditions for P.vulgaris triterpenoids were:ethanol concentration 62%,ultra-sound power 400 W,and ultrasound temperature 36℃.Under these conditions,the extraction yield of triterpenoids from P.vulgaris reached 23.16 mg/g.The in vitro hypoglycemic and anti-inflammatory activities evaluation results showed that the hypoglycemic activity of P.vulgaris triterpenoids increased with the concentration in the range of 0.5 mg/mL to 7 mg/mL.At a concentration of 7 mg/mL,the inhibition rate against α-glucosidase was 82.34%,and a-gainst α-amylase was 89.50%.The extraction process of triterpenoids from P.vulgaris were optimized and the hypo-glycemic activity was evaluated,which could provide a scientific basis for further development and utilization of P.vul-garis triterpenoids.
