Objective To investigate the effects of peroxisome proliferator-activated receptor γ(PPARγ)on the cognitive function of rats after propofol anesthesia.Methods Eighty rats were randomly divided into the control group(C)(with 10 mL/kg intraperitoneal injection of normal saline),the pioglitazone(P)group(with 10 mg/kg intraperitoneal injection of pioglitazone dissolved in normal saline),normal saline(N)group(with 10 mL/kg intraperitoneal injection of normal saline)and GW9662(G)group(with 100 mg of GW9662 inhibitor dissolved in dimethyl sulfoxide 100 mL,2 mg/kg intraperitoneal injection),and such performance lasted for 5 consecutive days.On day 6,exploratory laparotomy was performed under propofol anesthesia in Groups P,N,and G,while neither operation nor anesthesia was performed in Group C.Eight rats in each group were killed in anesthesia 24 h after operation,while the levels of amyloid beta(Aβ),phosphorylated microtubule-associated protein(P-Tau)and PPARγ in the hippocampus were detected by ELISA and Western blot,respectively.On the 5th day after surgery,two rats of each group were killed in anesthesia,while the hippocampal tissue was sectured and hematoxylin-eosin(HE)staining was used to observe the changes of hippocampal cells in each group.On the 7th day after surgery,the postoperative cognitive function of 10 rats from each group were detected by Morris water maze test.Results The deposition of Aβ and the expression of P-Tau in hippocampus of Group N increased compared with Group C(P<0.05),and decreased compared with Group N(P<0.05),and increased in Group G compared with Group N(P<0.05).The expression of PPARγ in hippocampus of Group P increased compared with that in Group N(P<0.05),and that in Group G decreased compared with that in Group N(P<0.05).The hippocampal cells of Groups C and P were relatively closely arranged and their membranes were clear,while the hippocampal cells of Groups G and N were disordered and their nuclei were pyknotic.Compared with Group C,the escape latency of Group N was prolonged,the number of crossing platform was reduced,and the swimming time of target quadrant was shorter(P<0.05).Compared with group N,group P had shorter escape incubation period,more times of platform crossing and longer swimming time in target quadrant(P<0.05),while group G had longer escape incubation period,less times of platform crossing and shorter swimming time in target quadrant(P<0.05).Conclusions The increase of PPARγ level can improve the cognitive dysfunction of rats after propofol anesthesia.
