Objective To investigate the expression of histone acetyltransferase P300 and deacetylase SIRT2 in breast invasive ductal carcinoma,and its relationship with clinicopathological features.Methods A total of 385 cases of breast invasive ductal carcinoma tissues as case group,36 precancerous tissues as control group.The tissue microarray was prepared,and the protein expression of P300 and SIRT2 in both groups was detected by immunohistochemical En Vision two-step method and then compared the expression differences.The relationship between P300 and SIRT2 protein and clinicopathological features of breast cancer patients was analyzed.The correlation between P300 and SIRT2 protein in patients with breast cancer was analyzed by Spearman method.Kaplan-Meier and Log rank test statistical methods were used to analyze the correlation between P300 and SIRT2 proteins and the prognosis of breast cancer patients.Results The low expression rate of P300 protein in the case group was higher than that of control group,and the low expression rate of SIRT2 protein was lower than that of control group,the difference was statistically significant(P<0.05).In breast cancer tissues,the low expression rate of P300 protein in tumor with maximum diameter>5 cm group was significantly higher than that of ≤5 cm group,the low expression rate in TNM stage Ⅲ-Ⅳ group was higher than that of stage Ⅰ-Ⅱ group,the low expression rate in lymph node metastasis group was higher than that of non-lymph node metastasis group,the low expression rate in Ki67 high expression group was higher than that of Ki67 low expression group,and the low expression rate in Luminal B type was higher than that of TNBC,the differences were statistically significant(P<0.05).The low expression rate of SIRT2 protein in histological 3 grading was lower than that of grade 1-2;the low expression rate in tumor with maximum diameter>5 cm group was higher than that of≤5 cm group,the low expression rate in Ki67 high expression group was lower than that of Ki67 low expression group,and the low expression rate in Luminal A type and Luminal B type was lower than that of HER2 positive type and TNBC,the differences were statistically significant(P<0.05).There was a negative correlation between the expression of P300 and SIRT2 protein(r=-1.77,P<0.001).The overall disease-free survival rate of the P300 protein low expression group was lower than that of the high expression group(P=0.046).Conclusions P300 and SIRT2 proteins might be related to the occurrence and development of breast invasive ductal carcinoma.Low expression of P300 protein might indicate poor prognosis in patients with breast invasive ductal carcinoma.
