Objective To investigate the expression of absent of melunoma 2(AIM2)inflammasome in eosinophilic chronic rhinosinusitis with nasal polyps(ECRSwNP)and its clinical significance.Methods Sixty patients who underwent endoscopic sinus surgery were divided into three groups:Control group(simple nasal septum deviation,n=19),non-ECRSwNP group(n=18),and ECRSwNP group(n=23).Gender,age,visual analogue scale(VAS)score,sinus Lund Mackay CT score and Lund-Kennedy nasal endoscopic score were compared among three groups.Reverse transcription quantitative PCR(RT-qPCR)was applied to detect AIM2 expression in the normal inferior turbinate of Control group,nasal polyps in non-ECRSwNP and ECRSwNP groups.EnVision,a two-step immunohistochemistry(IHC)staining technique,was performed to detect the expressions of AIM2,caspase-1,interleukin-1β(IL-1β),and IL-18 in corresponding tissues.Spearman correlation was analyzed between AIM2 and clinical symptoms of ECRSwNP.Results There were no statistically significant differences in gender and age among three groups(P>0.05),while there were statistically significant differences in VAS score,sinus Lund Mackay CT score and Lund Kennedy nasal endoscopic score among the groups(P<0.05).The expressions of AIM2,caspase-1,IL-1β,and IL-18 were significantly higher in ECRSwNP group than those in non-ECRSwNP and control groups(P<0.05).AIM2expression level in ECRSwNP group was positively correlated with VAS score(rs=0.649,P<0.05),sinus Lund Mackay CT score(rs=0.659,P<0.05),Lund Mackay nasal endoscopic score(rs=0.592,P<0.05)and the percentage of eosinophils in tissues(rs=0.584,P<0.05).Conclusion AIM2 inflammasome is highly expressed in ECRSwNP and associated with the severity of clinical symptoms of ECRSwNP.
