Research of HPA-1-6/10/15/21 and HLA-A/B gene polymorphism in platelet donors of Guiyang
Objective To study the gene distribution and polymorphism of human platelet antigen HPA-1-6/10/15/21 and human leukocyte antigens HLA-A and B from human-collected platelet donors in Guiyang.Methods The real-time fluorescent quantitative PCR method(qPCR)on 287 guiyang region machine adopt platelet donors in HPA-1-6/10/15/21 and HLA-A,B genotyping,The distribution of aa\ab\bb gene and HLA-A and B alleles were listed,and the frequencies of aa\ab\bb gene and HLA-A and B alleles were calculated.Results In the HPA-1-6/10/15/21 system of 287 platelet donors,the genotypes of HPA-4 and HPA-10 were both aa type and were not polymorphic.HPA-1,2,5,6 and 21 are mainly aa type,and the bb-type was detected only in HPA-3 and HPA-15.The highest heterozygosity was found in HPA-15,followed by the second HPA 3 heterozygosity.14 alleles were detected at the HLA-A site,and the three highest frequencies were A*02(0.36),A*11(0.33),and A*24(0.162).Twenty-three alleles were detected at the HLA-B locus,and the four highest frequencies were B*46(0.19),B*15(0.149),B*40(0.14),and B*13(0.13).Conclusion There are polymorphisms in HPA-1-6/10/15/21 and HLA-A and B genes of platelet donors in Guiyang,so it is necessary to establish a local HPA/HLA genotyping platelet donor bank for clinical use.
