Objective To investigate the expression and significance of microRNA-21(miR-21)and Forkhead Box Protein M1(FOXM1)in patients with Gestational Hypertension(GHT)and Preeclampsia(PE).Methods A total of 150 patients with hypertensive disorders of pregnancy(HDP)were enrolled,comprising 112 in the GHT group and 38 in the PE group,alongside 150 control subjects matched by age and gestational age.On the day of enrollment,4 mL of fasting peripheral venous blood was collected from all participants to measure serum miR-21 and FOXM1 levels.The correlation between serum miR-21 and FOXM1 mRNA in the GHT and PE groups were analyzed,the receiver operating characteristic(ROC)curve was drew,and the area under the curve(AUC)to evaluate factors influencing PE was calculated.It were also assessed the predictive value of miR-21 and FOXM1 mRNA for PE and their impact on pregnancy outcomes.Results Serum miR-21 and FOXM1 mRNA levels were significantly lower in the PE and GHT groups compared to controls,with the lowest levels observed in the PE group(P<0.05).Both groups showed a positive correlation between miR-21 and FOXM1 mRNA(P<0.05).MiR-21 and FOXM1 mRNA served as independent protective factors against PE(P<0.05).A risk model based on miR-21 and FOXM1 mRNA yielded an AUC of 0.841 for assessing PE,surpassing the AUCs when assessed individually(P<0.05).The PE group exhibited a higher incidence of adverse pregnancy outcomes compared to the GHT group(x2=4.053,P=0.044),with affected patients showing lower serum levels of miR-21 and FOXM1 mRNA than those with normal outcomes(P<0.05).Conclusion Decreased miR-21 expression may lead to the downregulation of FOXM1 mRNA,contributing to the development of GHT and PE and increasing the risk of adverse pregnancy outcomes.
