Expression of SIRT2,P65,and Survivin in breast cancer and their association with clinicopathological features
Objective To investigate the relationship between SIRT2,P65,and Survivin in breast cancer by detecting the expressions of SIRT2,P65,and Survivin,and explore their association with clinicopathological features.Methods A total of 384 patients with invasive breast cancer were selected as breast cancer group and 31 patients with fibroadenosis as control group.Immuno-histochemical EnVision two-step method was used to detect the protein expressions of SIRT2,P65,and Survivin in breast lesions in two groups.At the same time,clinicopathological features-related information of breast cancer group was collected,including age,molecular typing,TNM staging,histological grading,lymph node metastasis and menstruation.The association of SIRT2,P65,and Survivin protein expressions in breast cancer group with clinicopathological features were analyzed.The correlation between SIRT2,P65,and Survivin protein expressions in breast cancer group were analyzed.Results SIRT2 and Survivin proteins were highly expressed in breast cancer tissues.The rates of highly expressed SIRT2 and Survivin was higher in breast cancer group than those in control group(P<0.05).The protein expressions of SIRT2,P65,and Survivin in different molecular types and TNM stages were higher in breast cancer group than those in control group(P<0.05).The rate of highly expressed SIRT2 protein was higher in Luminal B1 than that in other types,while the rate of highly expressed P65 protein was higher in Luminal A than that in other types.The rate of highly expressed SIRT2 protein was higher in HER2+breast cancer than that in other types,while the rates of highly expressed SIRT2,P65,and Survivin proteins were higher in TNM Ⅲ~Ⅳ than those in TNMⅠ-Ⅱ.SIRT2 and P65 protein expressions were higher in the patients with lymph node metastasis of breast cancer than those in the patients with non-lymph node metastasis(P<0.05),and SIRT2-and P65-positive rates of the samples with their high expressions were higher than those in SIRT2-and P65-negative rates(P<0.05).P65 and Survivin proteins were highly expressed in histologically graded breast cancer patients(P<0.05),and the rates of highly expressed P65 and Survivin proteins were higher in grade Ⅰ than those in grade Ⅱ/Ⅲ.In breast cancer group,SIRT2 protein expression was positively correlated with P65 and Survivin protein expression(r=0.371 and 0.310,P<0.001,respectively),and P65 was positively correlated with Survivin protein expression(r=0.466,P<0.001).Conclusion The expression of SIRT2 and Survivin protein are upregulated in breast cancer tissues,which may be the mechanism influencing the clinicopathological characteristics of breast cancer,such as histological grading,TNM staging and molecular typing.
