Box-Behnken响应面法优化青钱柳黄酮类成分提取工艺

通过试验优选出青钱柳黄酮类成分的最佳提取工艺参数.使用高效液相色谱法,以青钱柳中山柰酚-3-O-β-D-葡萄糖醛酸苷、槲皮素-3-O-β-D-葡萄糖醛酸苷、金丝桃苷、阿福豆苷的总提取率作为评价指标,并且分别对加热回流时间、料液比、甲醇体积分数进行单因素试验,随后采用Box-Behnken响应面法试验对工艺参数进行优化,优选出青钱柳黄酮类成分的最佳提取工艺.试验结果表明青钱柳黄酮类成分的最佳提取工艺参数为 75%的甲醇,1∶55 的液料比,加热回流 47 min,提取率为2.04%,试验结果与预测值接近.优选出的工艺参数合理、可靠、可操作性高,可作为工业化提取青钱柳黄酮类成分的参考依据.
Optimization on Extraction Process of Flavonoids from Cyclocarya Paliurus by Box-Benhnken Response Surface Methodology
The optimum extraction parameters of flavonoids from Cyclocarya paliurus were obtained through experiments.The total extraction rate of Kaempferol-3-O-β-D-glucuronide,Quercetin-3-O-β-D-glucuronide,Hyperoside and Afzelin from Cyclocarya paliurus was evaluated using high-performance liquid chromatography(HPLC),and a one-factor test was conducted for heating and refluxing time,material-liquid ratio,and volume fraction of methanol,respectively,subsequently,the Box-Behnken response surface method test was used to optimize the process parameters,and the best extraction process for the flavonoid components of Cyclocarya paliurus was preferred.The experimental results showed that the optimal extraction process parameters for the flavonoid components of Cyclocarya paliurus were 75%methanol,1∶55 liquid to material ratio,heating and refluxing for 47 min,with an extraction rate of 2.04%,and the experimental results were close to the predicted values.The preferred process parameters were reasonable,reliable and highly operable,which could be used as the reference basis for the industrialized extraction of the flavonoid components from Cyclocarya paliurus.
