Optimization on Extraction Process of Flavonoids from Cyclocarya Paliurus by Box-Benhnken Response Surface Methodology
The optimum extraction parameters of flavonoids from Cyclocarya paliurus were obtained through experiments.The total extraction rate of Kaempferol-3-O-β-D-glucuronide,Quercetin-3-O-β-D-glucuronide,Hyperoside and Afzelin from Cyclocarya paliurus was evaluated using high-performance liquid chromatography(HPLC),and a one-factor test was conducted for heating and refluxing time,material-liquid ratio,and volume fraction of methanol,respectively,subsequently,the Box-Behnken response surface method test was used to optimize the process parameters,and the best extraction process for the flavonoid components of Cyclocarya paliurus was preferred.The experimental results showed that the optimal extraction process parameters for the flavonoid components of Cyclocarya paliurus were 75%methanol,1∶55 liquid to material ratio,heating and refluxing for 47 min,with an extraction rate of 2.04%,and the experimental results were close to the predicted values.The preferred process parameters were reasonable,reliable and highly operable,which could be used as the reference basis for the industrialized extraction of the flavonoid components from Cyclocarya paliurus.
