Study on Extraction Process of Licorice Based on Bacteriostatic Experiment and Orthogonal Optimization
Licorice is a traditional medicine and food homologous substance.It not only has many effects such as tonifying spleen and replenishing qi,clearing heat and detoxifying,but also has good antibacterial activity.At present,most of the studies are based on the content of one or several active ingredients to optimize the extraction process,and often cannot obtain a process with good antibacterial activity.Based on the inhibition zone diameter of oral pathogenic bacteria Streptococcus mutans and Porphyromonas gingivalis,the comprehensive bacteriostatic score was used as the optimization index.The extraction process of licorice water extract was optimized by single factor experiment and orthogonal experiment on the amount of water added,the number of water added and the extraction time.The single factor experiment showed that the best amount of water was 10 times,the extraction times were 3 times,and the extraction time was 2 h.The orthogonal experiment showed that the water extract of licorice obtained by extracting 3 times with 8 times the amount of water and extracting 2 h each time had the best comprehensive score for the two oral pathogens.Based on the antibacterial activity of two kinds of oral pathogenic bacteria,the extraction process of licorice with high resistance to oral pathogenic bacteria was obtained by single factor and orthogonal experiments,which provided data support for the development of licorice in the prevention or treatment of oral pathogenic bacteria infection.
