目的:建立高效液相色谱法测定绿豆衣中牡荆素和异牡荆素含量.方法:采用高效液相色谱法(HPLC)测定.色谱柱为Thermo C18(4.6 mm×250 mm,5 μm),流动相为甲醇-0.5%磷酸(40∶60,V/V),流速为1.0 mL/min;检测波长为340 nm,进样体积10 μL.结果:牡荆素和异牡荆素质量浓度分别在10.0~80.0 μg/mL和2.00~30.0 μg/mL范围内与峰面积线性关系良好(r=0.9999 和r=0.9999);平均加样回收率分别为 98.89%(RSD=0.21%)和 99.39%(RSD=0.26%).结论:建立的HPLC方法简便、准确、灵敏度高、专属性强,可用于绿豆衣中牡荆素和异牡荆素的含量测定.
Determination of two constituents in mung bean hull by HPLC
In this study we established an HPLC method for the content determination of vitexin and isovitexin in mung bean hull.The Thermo C18 column(4.6 mm×250 mm,5 μm)was adopted,the mobile phase was methanol-0.5%phosphoric acid(40∶60,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 340 nm,and the injection volume was 10 μL.Vitexin and isovitexin had good linear relationship in the concentration ranges of 10.0-80.0 μg/mL and 2.00-30.0 μg/mL(r=0.9999),the average spiked recoveries were 98.89%and 99.39%,and the RSD were 0.21%and 0.26%.The established HPLC method is simple,accurate,sensitive and specific,and can be used for the content determination of vitexin and isovitexin in mung bean hull.
