芪锦颗粒提取纯化工艺研究

目的:优选芪锦颗粒的提取纯化工艺.方法:以出膏率、莫诺苷含量为评价指标,通过单因素试验和L9(34)正交试验,综合评价制定芪锦颗粒提取工艺,参照处方中各有效成分的理化性质,拟定出以乙醇为溶剂进行澄清工艺研究,确定最佳的提取纯化工艺条件.结果:最佳提取纯化条件为饮片8倍量的水,煎煮提取3次,每次0.5 h,合并提取液浓缩至相对密度为1.23~1.28的浓缩液,缓慢加入95%的乙醇,边加边搅拌,使其醇浓度达80%为止,静止过夜,过滤,滤液回收乙醇后备用.醇沉后出膏率为19.5%,莫诺苷提取率为93.50%.最大限度地保留了制剂有效成分.结论:该提取方法可行,纯化条件下有效成分保留率高、除杂效果好,适用于芪锦颗粒提取纯化工艺研究.
Study on extraction and purification technology of Qijin Granules
We optimized the extraction and purification process of Qijin Granules.The extraction technology of Qi-jin Granules was comprehensively evaluated by single factor tests and L9(34)orthogonal test with the extraction rate and morroniside content as evaluation indexes.The clarification process with ethanol as solvent was formulated ac-cording to the physicochemical properties of each active component in the prescription,and the optimal extraction and purification conditions were determined.The optimal extraction and purification conditions were as follows;8 times the amount of water;decocting and extracting 3 times,0.5 h each time;concentrating the combined extraction solution to the concentrated solution with a relative density of 1.23 to 1.28;adding 95%ethanol slowly while stir-ring,until the alcohol concentration reached 80;standing still overnight,filtrating and recovering the ethanol.The extraction rate was 19.5%,and the extraction rate of morroniside was 93.50%.The active ingredients of the prep-aration were retained to the maximum extent.The extraction method is feasible,has high retention rate of the effec-tive components and good effect of impurity removal,and is suitable for the extraction and purification process of Qi-jin Granules.
