脊蛇祛湿胶囊调节mPGES-1干预类风湿关节炎的作用机制研究

摘要
目的:探究脊蛇祛湿胶囊(JiShe QuShi Capsules,JSQSC)对胶原诱导性关节炎(Collagen-induced Arthritis,CIA)大鼠模型微粒体前列腺素E2 合酶-1(mPGES-1)靶点的影响.方法:雌性SD大鼠24 只,随机分为正常组、模型组、JSQSC组和甲氨蝶呤(Methotrexate,MTX)组,每组6 只.适应性喂养一周后构建CIA大鼠模型,二次免疫后第3 天开始给药.JSQSC组每天灌胃脊蛇祛湿胶囊药液,MTX组每天灌胃1 mg/kg甲氨蝶呤药液,正常组和模型组每天灌胃等体积的生理盐水,每 7 天检测大鼠体重、足肿胀等指标,于实验第28 天取材.结果:相比于模型组,JSQSC组与MTX组的大鼠体质量增加,足肿胀减轻,滑膜组织mPGES-1 表达水平降低(P<0.05).结论:脊蛇祛湿胶囊药液可通过抑制mPGES-1 的表达从而减轻CIA大鼠炎性症状的产生.
Abstract
We investigated the effects of Jishe Qushi Capsules(JSQSC)on microsomal prostaglandin E2 synthase-1(mPGES-1)targets in a rat model of collagen-induced arthritis(CIA).Twenty-four female SD rats were ran-domly divided into normal,model,JSQSC and methotrexate(MTX)groups,with six rats in each group.The CIA rat model was constructed after one week of acclimatization feeding,and the administration was started on the third day after secondary immunization.JSQSC solution was gavaged daily in the JSQSC group,1 mg/kg of methotrexate solu-tion was gavaged daily in the MTX group,and an equal volume of saline was gavaged daily in the normal and model groups.The body weight,foot swelling and other indexes of the rats were measured every 7 days,and the samples were taken on the 28th day of the experiment.Compared with the model group,the rats in the JSQSC and MTX groups had increased body mass,reduced foot swelling and decreased mPGES-1 expression level in synovial tissue(P<0.05).In conclusion,Jishe Qushi Capsules could reduce the inflammatory symptoms in CIA rats by inhibiting the expression of mPGES-1.
