三种市场流通茶菊的分子鉴定学研究

目的:为了更好地对市场上流通的茶菊品种进行准确鉴定,进一步规范茶菊市场的管理和监督.方法:利用分子鉴定的手段,选用4 个常用的DNA条形码片段(ITS、rbcL、psbA-trnH和matK)对市场上购买的金丝皇菊、婺源皇菊、杭白菊三个茶菊品种的不同产地样品进行分子鉴定学研究.结果:本研究共获得金丝皇菊、婺源皇菊及杭白菊序列 117 条.在各品种的鉴定方面,ITS片段的区分度相对而言最好,金丝皇菊的所有24 个样品在ITS构建的NJ树中可聚为一支.结论:ITS片段相对于rbcL和psbA-trnH具有更好的菊茶品种分辨率.但要想彻底理清菊茶各品种之间的系统进化关系,还需借助基因组水平的分子系统学研究.
Molecular identification of three varieties of tea chrysanthemum in the market
In order to accurately identify the tea chrysanthemum varieties in the market and further standardize the management and supervision of the tea chrysanthemum market,four common DNA barcode fragments(ITS,rbcL,psbA-trnH and matK)were selected to conduct molecular identification study on three tea chrysanthemum varieties from different origins.A total of 117 sequences were obtained from the three tea chrysanthemum varieties.The ITS fragment had the best discrimination.All 24 samples of Jinsi chrysanthemum clustered together in the NJ tree con-structed with ITS.In conclusion,compared with rbcL and psbA-trnH,ITS fragment has better resolution of tea chrys-anthemum varieties.However,in order to thoroughly clarify the phylogenetic relationship between various tea chrys-anthemum varieties,molecular systematic study at the genomic level is needed.
