Regeneration System Establishment of Tissue Culture of Colacasia escuenla in Hezhou
The effects of disinfection time and ectohormone on the survival rate, induction,multiplication and differentiation of callus and root formation were studied by taking young leaf of Colacasia escuenla .in Hezhou as explants. The results showed that the pollution rate and browning rate decreased when treated with 50% calcium hypochlorite for 6 min, and the survival rate was 81%. The dominant factors for callus induction were in the order of KT, IBA and BA, the optimum induction medium was MS+IBA 0.1 mg/L+KT 1.0mg/L+BA 1.0mg/L and the average induction rate was 99%;the optimum medium for multiplication and differentiation was MS+ KT 3.0mg/L+ IBA 0.5 mg/L and the multiplication coefficient was 3.0. Adding 3 g/L yeast extract to the medium for multiplication could significantly increase seedling height growth and dry matter weight. The best rooting medium was 1/2 MS+NAA 0.5 mg/L, the rooting rate was 100%, the survival rate was high with thick and strong lateral roots and dense fibrous roots.
