为了考察三叶木通叶片保存时间与基因组DNA提取质量的关系,以低温保存1个月扣6个月的三叶木通[Akebia trifoliata( Thunb) Koidz]叶片为材料,采用改良的CTAB法提取其基因组DNA,用琼脂糖凝胶电泳和紫外分光光度计检测获得的DNA质量.结果表明:低温保存1个月三叶木通叶片中提取的DNA质量较高,检测条带清晰,可用于后期的分子生物学研究.结论:三叶木通叶片保存时间过长,其核酸结构易发生改变而降解,从而影响DNA的提取质量.
With the purpose of studying the effect of preservation time to the extraction of genomic DNA from A. Trifoliate, the genome DNA was isolated by improved CTAB method with the leaves that preserved for one month and six months under low temperature respectively. The quality of the DNA was examined with agarose gel electrophocesis and ultraviolet sepectrophotometer. The results showed the high quality DNA was extracted from the leaves that preserved for one month, which could be used for the next study. So the leaf would disintegrate if preserved for too long a time, and the extraction of DNA would be affected.
