产抗凝溶栓双活性物质菌株ZLX 3和MJM 38的筛选及鉴定

为筛选可产生抗血栓活性物质的Chitinophaga菌株,利用 VY/4平板法从兔粪、羊粪、土样、朽木等样品中筛选目的菌株,纤维蛋白平板法检测溶栓活性,纤维蛋白试管法检测抗凝活性,通过形态学特征、理化性质、G+C含量、16 S rRNA序列同源性鉴定目的菌株。结果表明:从朽木样品中分离到可产生抗血栓活性物质的菌株 ZLX 3和菌株 MJM 38,两者分泌的胞外液均有明显的溶栓活性和抗凝活性,均属于Chitinophaga菌属,其中菌株 ZLX 3属于种 Chitinophaga pinensis ,将其命名为 Chitinophaga pinensis ZLX 3,菌株 MJM 38属于种Chitinophagarupis ,将其命名为 Chitinophagarupis MJM 38。结论:菌株Chitinophaga pinensis ZLX 3和菌株Chitinophagarupis MJM 38均可产生抗凝溶栓双活性物质。
Screening and Identification of Chitinophaga pinensis ZLX 3 and Chitinophaga rupis sp.MJM 38 Producing Anticoagulation and Thrombolysis Double Active Substance
To screen the Chitinophaga strains that can produce the antithrombotic active substance, the authors screened the target strains on VY/4 plate from dozens of samples such as rabbit manure,sheep manure,soil,deadwood,etc.and detected the fibrinolytic activity of the target strains by the fibrin plate and the anticoagulant activity by fibrin tube, then identified the target strains by morphological characteristics,physical and chemical properties and G+C content of DNA and 16S rRNA sequence homology.Results:the strains ZLX 3 and MJM 38 that can produce antithrombotic active substance were obtained from the rotten wood samples.And found that the extracellular fluid of the strains ZLX 3 and MJM 38 showed both obvious fibrinolytic activity and obvious anticoagulation activity.The strain ZLX 3 belonged to the Chitinophaga pinensis and the strain MJM 38 belonged to the Chitinophaga rupis.The strains ZLX 3 was named as Chitinophaga pinensis ZLX 3 and the strains MJM 38 as Chitinophaga rupis sp.MJM 38. Conclusion:Both the strains ZLX 3 and the strains MJM 38 can produce obvious anticoagulation and thrombolysis double active substance.
