Solid State Fermentation of Xylanase from Aspergillus niger and Property of Crude Enzyme
The solid state fermentation condition of xylanase from A.niger XZ-3S strain and property of the produced crude enzyme were studied and determined by tray culture and enzyme activity method. The results showed that the xylanase activity can reach 24 380 IU/g under the fermentation conditions of 100g dry substance,1 ∶ 1.7 of dry substance :water,natural initial pH,10% inoculation size of dry substance,timing stirring and watering and 28℃ for 60h,and the optimal pH and temperature for the produced crude enzyme is 5.0 and 50℃ respectively.The property of the crude enzyme is stable under the conditions of below 50℃ and pH3.0~8.0.EDTA,Cu2+ and Co2+ have the obvious activation effect on activity of xylanase but Pb2+ ,Mg2+ ,Fe2+ ,Mn2+ ,Ca2+ ,Fe3+ and Al3+ have different inhibition effect on activity of xylanase to varying degrees respectively.
