Extraction and Stability of Chlorophyll from Itea ilicifolia Leaves
The chlorophyll samples extracted from Ⅰ.ilicifolia leaves by using seven different extraction solvents under room temperature and ice-bath condition,and then the effect of different illumination time,different temperature,different treatment time of H2O2with different concentration and different metal ions on stability of chlorophyll samples was studied to provide a theoretical reference for physiological and ecological studies of Ⅰ.ilicifolia.Results:The absorbance of chlorophyll a and chlorophyll b extracted from Ⅰ.ilicifolia by the mixed solvent of absolute ethyl alcohol and acetone (1 ∶1) at room temperature is 1.622 and 0.675 respectively but the absorbance of chlorophyll a and chlorophyll b extracted from Ⅰ.ilicifolia by 80% acetone is 0.458 and 0.264 separately,which indicates that the mixed solvent of absolute ethyl alcohol and acetone (1 ∶ 1) is the best extraction solvent and the 80% acetone solvent is the worst extraction solvent for extracting chlorophyll from Ⅰ.ilicifolia.H2O2 has a large influence on stability of chlorophyll extracted from Ⅰ.ilicifolia but temperature has a little influence on stability of chlorophyll extracted from Ⅰ.ilicifolia.Illumination time has a large influence on stability of chlorophyll a extracted from Ⅰ.ilicifolia.K+,Na+,Zn2+ and Mg2+ have no obvious influence on stability of chlorophyll a extracted from Ⅰ.ilicifolia.But Al3+,Fe2+,Cu2+ and Ca2+ can reduce stability of chlorophyll a extracted from Ⅰ.ilicifolia.
