Development of Microsatellite in Cranoglanis bouderius by Magnetic Beads
To provide the theoretical basis for genetic breeding,germplasm identification and releasing.Magnetic beads enriched method was used to isolate microsatellites from C.bouderius,through sequencing and analysis the positive colonies,the repeats which contained enough flanking region were obtained to design 32 microsatellite primers by the software Primer Premier 5.0.Results:80 positive colonies were obtained through 596 white spots.Sequencing of the 80 positive colonies confirmed that 47 microsatellite sequences were obtained.From these sequences,31 sequences(66 %) were perfect repeats,14(30%) were imperfect repeats,and two sequences (4%)were compound repeats.27 pairs of microsatellite primers used successfully to amplify special fragments at appropriate annealing temperature,and 24 pairs of them were polymorphism within species.
