Genetic Diversity of Chrysanthemum Germplasm Resources in Yantai by Using STR Fluorescence Markers
[Objective]The genetic diversity of chrysanthemum germplasm resources in Yantai area was studied,which supported the classification,resource protection,and variety cultivation of chrysanthemum germplasm in Yantai area.[Method]Fluorescent STR molecular markers and capillary electrophoresis were used to analyze 12 chrysanthemum germplasms from Yantai area,and cluster analysis was conducted based on the observation of major phenotypic traits to construct molecular ID.[Result]A total of 103 STR alleles were detected from 10 pairs of primers,with an average of 10.3 alleles per primer pair.The lengths of the amplified fragment were 118-376 bp,and the primer polymorphism information(PIC)averaged 0.795 8.Cluster analysis of chrysanthemum germplasm was conducted,and 12 germplasm were divided into 4 categories based on genetic similarity coefficient.Group Ⅰ included Chrysanthemum morifolium'Qiuju',Bellis perennis,Chrysanthemum indicum(1),Chrysanthemum mori folium'Hangju'(white),Chrysanthemum morifolium'Hangju'(yellow),Callistephus chinensis and Chrysanthemum indicum(2),which accounted for 58.3%of 12 chrysanthemum germplasms,including 1 large-flowered chrysanthemum,6 small-flowered chrysanthemums,2 flat-type petals,3 spoon-type petals,2 tubular type petals.Group Ⅱ included Gerbera jamesonii and Matricaria recutita,which accounted for 16.7%of total chrysanthemums,and their petals were all flat type.Only Chrysanthemum morifolium'Hangju'(purple)belonged to Group Ⅲ,which accounted for 8.3%of total chrysanthemums.Group Ⅳ included Erigeron annuus and Chrysanthemum indicum(3),accounting for 16.7%,and the two cultivars were both small-flowered chrysanthemums with flat-type petals.There was cross and aggregation in different flower color,inflorescence size and petal type.[Conclusion]Chrysanthemum germplasm in Yantai area has good genetic diversity,and STR fluorescence markers can effectively analyze the type of germplasm resources.The molecular ID of 12 chrysanthemum germplams were constructed based on STR molecular method.
